microRNA-145对癫痫细胞炎症反应的影响

目的 探讨microRNA(miR)-145 对癫痫细胞炎症反应的影响。方法 体外培养经白细胞介素-1β(interleukin-1β,IL-1β)处理过的CTX-TNA2细胞24 h,随机均分为NC组(不转染)、inhibitor NC组(转染无义序列inhibitor)、miR-145 inhibitor组(转染miR-145 inhibitor)、mimics NC组(转染无义序列mimics)、miR-145 mimics组(转染miR-145 mimics)。CCK8 检测各组细胞活力,应用定量即时聚合酶链锁反应(quantitative real time polymerase chain reaction,RT-qPCR)和酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA)检测各组细胞炎症因子白细胞介素-2(interleukin-2,IL-2)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)mRNA和蛋白表达水平。结果 NC组IL-2、IL-6、TNF-α 蛋白表达分别为(532。77±5。33)pg/L、(55。48±0。79)pg/L、(266。85±6。61)ng/mL;miR-145 mimics组IL-2、IL-6、TNF-α蛋白表达分别为(622。24±9。66)pg/L、(76。59±0。98)pg/L、(359。94±2。20)ng/mL,与NC组比较,miR-145 mimics组IL-2、IL-6、TNF-α的表达水平均升高(P<0。01);经miR-145 抑制剂干预后,IL-2、IL-6、TNF-α的表达水平均较NC组和miR-145 mimics组下降(P<0。01)。在 12、24、48 h,与NC组、miR-145 inhibitor组比较,miR-145 mimics组细胞活力均下降(P<0。01);与NC组、miR-145 mimics组比较,miR-145 inhibitor组细胞活力均升高(P<0。01)。结论 miR-145 可调节癫痫细胞炎症反应过程,抑制miR-145 可抑制炎症反应。
The Effect of microRNA-145 on the Inflammatory Response of Epileptic Cells
Objective To investigate the effect of microRNA(miR)-145 on the inflammatory response of epileptic cells.Methods CTX-TNA2 cells treated with interleukin-1β(IL-1β)were cultured in vitro for 24 h.They were randomly divided into NC group(non-transfection),inhibitor NC group(transfection nonsense sequence inhibitor),miR-145 inhibitor group(transfection miR-145 inhibitor),mimics NC group(transfection nonsense sequence mimics)and miR-145 mimics group(transfected with miR-145 mimics).CCK8 was used to detect cell viability in each group,and quantitative real time polymerase chain reaction(RT-qPCR)and enzyme-linked immunosorbent assay(ELISA)were used to detect inflammatory factors interleukin-2(IL-2),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)in each group mRNA and protein expression levels.Results The expressions of IL-2,IL-6 and TNF-α in NC group were(532.77±5.33)pg/L,(55.48±0.79)pg/L and(266.85±6.61)ng/mL,respectively.The expressions of IL-2,IL-6 and TNF-α in miR-145 mimics group were(622.24±9.66)pg/L,(76.59±0.98)pg/L,and(359.94±2.20)ng/mL,respectively.Compared with the NC group,the expression levels of IL-2,IL-6 and TNF-α in miR-145 mimics group were increased(P<0.01).After the intervention of miR-145 inhibitors,the expression levels of IL-2,IL-6 and TNF-α were decreased compared with those in NC group and miR-145 mimics group(P<0.01).At 12,24,48 h,compared with NC group and miR-145 inhibitor group,cell viability of miR-145 mimics group was decreased(P<0.01).Compared with NC group and miR-145 mimics group,the cell viability of miR-145 inhibitor group was increased(P<0.01).Conclusion miR-145 can regulate the inflammatory response process of epileptic cells,and inhibiting miR-145 can inhibit the inflammatory response.
