The Effect of microRNA-145 on the Inflammatory Response of Epileptic Cells
Objective To investigate the effect of microRNA(miR)-145 on the inflammatory response of epileptic cells.Methods CTX-TNA2 cells treated with interleukin-1β(IL-1β)were cultured in vitro for 24 h.They were randomly divided into NC group(non-transfection),inhibitor NC group(transfection nonsense sequence inhibitor),miR-145 inhibitor group(transfection miR-145 inhibitor),mimics NC group(transfection nonsense sequence mimics)and miR-145 mimics group(transfected with miR-145 mimics).CCK8 was used to detect cell viability in each group,and quantitative real time polymerase chain reaction(RT-qPCR)and enzyme-linked immunosorbent assay(ELISA)were used to detect inflammatory factors interleukin-2(IL-2),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)in each group mRNA and protein expression levels.Results The expressions of IL-2,IL-6 and TNF-α in NC group were(532.77±5.33)pg/L,(55.48±0.79)pg/L and(266.85±6.61)ng/mL,respectively.The expressions of IL-2,IL-6 and TNF-α in miR-145 mimics group were(622.24±9.66)pg/L,(76.59±0.98)pg/L,and(359.94±2.20)ng/mL,respectively.Compared with the NC group,the expression levels of IL-2,IL-6 and TNF-α in miR-145 mimics group were increased(P<0.01).After the intervention of miR-145 inhibitors,the expression levels of IL-2,IL-6 and TNF-α were decreased compared with those in NC group and miR-145 mimics group(P<0.01).At 12,24,48 h,compared with NC group and miR-145 inhibitor group,cell viability of miR-145 mimics group was decreased(P<0.01).Compared with NC group and miR-145 mimics group,the cell viability of miR-145 inhibitor group was increased(P<0.01).Conclusion miR-145 can regulate the inflammatory response process of epileptic cells,and inhibiting miR-145 can inhibit the inflammatory response.
