Establishment and Validation of a Method for Detecting Kk Blood Group Antigen Using a Fully Automated Blood Group Analysis System
Objective The non-supporting detection methods for Kk blood group antigen detection by BECKMAN PK7300 automatic blood group analysis system were established and verified,providing basic data support for routine Kk antigen detection in the later stage.Methods A total of 40 unpaid blood donors from Qingdao Blood Center from November 2021 to April 2022 were randomly selected.Based on the results of literature review,the experiment parameters were optimized to select incubation temperature,reagent dosage and dilution specimen dosage.According to the recommended parameters of the equipment manufacturer,the influence of the diluted concentration of red blood cells,anti-K and anti-K serum on the sample detection was evaluated.Results The parameters of Kk antigen were obtained by using PK7300 automatic blood group analyzer.The diluted concentrations of red blood cells and anti-K and anti-K serum were 1.50%and 1:20,respectively.The incubation temperature and time were 30℃and 1 h.The reaction volume of the reagent and sample was the same,both of which were 25 μL.Conclusion The experimental parameters of PK7300 non-matching blood group detection system established for Kk antigen detection have good specificity and high sensitivity,which are in line with the experimental expectation.The detection and verification of samples and reagent dilution ratio will help the detection system established in this study to accurately interpret the rare blood group batch detection and improve the detection efficiency,while reducing the loss of reagents.
