Comparison of Preparation Quality Between Whole Blood Collection and White Blood Cell Suspension on the Same Day and After Refrigeration for 24 Hours
Objective To compare the effects of whole blood collection day and refrigeration for 24 hours on the quality of preparing white blood cell suspension.Methods A total of 245 samples of 400 mL whole blood from the Qingdao Blood Center from January to October 2023 were selected and included in the observation group.The other 245 samples of 400 mL whole blood were included in the control group.The observation group was stored under 4℃refrigeration conditions for 24 hours before preparing white blood cell suspension.The control group underwent white blood cell suspension preparation on the day of collection.The filtration time,filter blood loss,cell count,metabolic indicators,free hemoglobin value,plasma hemolysis rate,and other quality indicators between two groups of white blood cell suspension were compared.Results There was no statistically significant difference in blood loss between the two groups of filters(P>0.05).The observation group had a larger volume of suspended red blood cells and a longer filtration time than the control group(P<0.05).There was no statistically significant difference in hemoglobin values and red blood cell counts between the two groups(P>0.05).The average red blood cell volume(90.97±4.79)fL in the observation group was greater than that(89.63±5.15)fL in the control group(P<0.05).There was no statistically significant difference in the levels of potassium ions,sodium ions,pH,and free hemoglobin content between the two groups(P>0.05).The plasma hemolysis rate in the observation group was 4.49%(11/245),which was higher than 1.22%(3/245)of the control group(P<0.05).Conclusion Compared with the day of whole blood collection,refrigeration for 24 hours has no significant effect on the loss of blood volume in the filter prepared by removing white suspended red blood cells.However,it increases the capacity of white suspended red blood cells,prolongs the filtration time,and has a certain impact on the cell count and plasma hemolysis rate detected.
