目的 探讨circRNA5430的表达对在肺腺癌细胞增殖和凋亡能力的影响。方法 通过慢病毒感染和嘌呤霉素药物筛选的方法构建稳定表达的NCI-H1975细胞株;通过实时定量逆转录聚合酶链反应(RT-PCR)检测细胞株中circRNA5430表达水平;Annexin V-APC和7-AAD双染法检测细胞凋亡水平;CCK-8法检测肺腺癌细胞增殖水平。结果 加入嘌呤霉素筛选14 d后可获得稳定表达circRNA5430的NCI-1975细胞株,实验组circRNA5430表达水平表达量较对照组明显升高;与对照细胞株相比,实验组细胞凋亡水平上升,2组间差异有统计学意义(P<0。001);与对照组相比,在24 h、48 h、72 h实验组的细胞增殖能力降低(P<0。05)。结论 高表达circRNA5430抑制肺腺癌细胞株增殖、促进肺腺癌细胞株凋亡。
Effect of high expression circRNA5430 on proliferation and apoptosis of lung adenocarcinoma cell line
Objective This paper aims to investigate the effect of circRNA5430 expression on the proliferation and apoptosis in lung adenocarcinoma cells(LAC).Methods The stable expression of NCI-H1975 cell line was constructed by lentivirus infection and puromycin drug screening.The expression of circRNA5430 was detected by qRT-PCR.Apoptosis was detected by Annexin V-APC and 7-AAD double staining.The proliferation of lung adenocarcinoma cells was detected by CCK-8 method.Results The NCI-H1975 cell line with stable expression of circRNA5430 was obtained after 14 days of screening with puromycin.The level of circRNA5430 in the experimental group was higher than that in the control group.Compared with the control cell line,the level of apoptosis in the experimental group increased,and the difference between the two groups was statistically significant(P<0.001).Compared with the control group,the cell proliferation ability of the experimental group decreased at 24 h,48 h and 72 h(P<0.05).Conclusion The overexpression of circRNA5430 inhibits the proliferation of lung adenocarcinoma cell lines and promotes the apoptosis of lung adenocarcinoma cell lines.
