桃红四物汤对放射性肠炎小鼠肠道微生物多样性的影响

目的 探究桃红四物汤对放射性肠炎小鼠的疗效,并初步解析其潜在肠道微生物相关作用机制。方法 将36只雄性SPF级C57BL/6J小鼠随机分为4组,即健康对照组(CON组)、放射性肠炎模型组(MODEL组)、桃红四物汤高剂量治疗组(THSWT_H组)和桃红四物汤低剂量治疗组(THSWT_L组)。除CON组小鼠只进行麻醉处理外,其余各组小鼠在麻醉状态下进行10Gy的全腹部辐射以建立放射性肠炎模型,并在指定时间点灌胃不同浓度的桃红四物汤,CON组和MODEL组小鼠灌胃等体积的生理盐水。实验终点时,采集血清、结肠组织和结肠内容物分别进行H&E染色、ELISA检测和16SrRNA扩增子测序,以评价桃红四物汤对小鼠炎症状态和血管功能、组织病理学损伤及肠道微生物的影响。结果 桃红四物汤可有效缓解放射性肠炎小鼠的全身性炎症反应,不同浓度桃红四物汤干预组小鼠血清中TNF-α、TGF-β、IL-6和IL-22水平均较MODEL组小鼠显著降低,而不同浓度桃红四物汤干预组小鼠血清中IL-10水平较MODEL组显著升高。血管功能生物标志物VEGF和Ang Ⅰ水平也发现类似的结果。H&E染色结果显示,桃红四物汤可改善辐射导致的小鼠结肠组织损伤。此外,不同浓度的桃红四物汤干预能有效改善放射性肠炎小鼠的肠道微生物紊乱,表现为不同浓度的桃红四物汤干预后,高、低剂量治疗组小鼠肠道微生物alpha多样性均高于MODEL组,且beta多样性均与CON组更接近。结论 桃红四物汤对放射性肠炎小鼠有较好的治疗效果,并能显著改善因辐射暴露造成的肠道微生物紊乱以及血管功能损伤。
The effect of Taohong Siwu Decoction on gut microbial diversity in mice with radiation-induced enteritis
Objective To observe the efficacy of Taohong Siwu Decoction(THSWD)on radiation-induced enteritis(RIE)in mice,and the probable mechanisms of action associated with gut microbiota.Methods Thirty six male SPF C57BL/6J mice were randomly divided into four groups:healthy control group(CON group),radiation enteritis model group(MODEL group),high-dose treatment group with THSWD(THSWD_H group),and low-dose treatment group with THSWD(THSWD_L group).Except for the CON group mice,which were only anesthetized,all the other groups of mice were subjected to 10 Gy of full abdominal radiation under anesthesia to establish the RIE models.Different concentrations of THSWD were orally administered at designated time points,while the CON and MODEL groups of mice were orally ad-ministered an equal volume of physiological saline.At the end of the experiment,the specimens of serum,colon tissue and colon contents were collected for H&E staining,ELISA detection,and 16S rRNA amplicon sequencing,respectively,to evaluate the effects of THSWD on inflammation status,vascular function,histopathological damage,and gut microbiota in mice.Results THSWD successfully reduced systemic inflammatory responses in RIE mice.Additionally,the serum levels of TNF-α,TGF-β,IL-6 and IL-22 in the intervention groups with varying dosages of THSWD were significantly de-creased,while that of IL-10 were significantly increased compared to the MODEL group respectively.Comparable out-comes were seen for the indicators of vascular function,VEGF and Ang Ⅰ.THSWD might ameliorate radiation-induced colon tissue damage in mice,according to the results of the H&E staining.Furthermore,THSWD intervention at varying concentrations could effectively improve the gut microbial disorder in mice with RIE.This was demonstrated by the gut microbiota of these mice at varying concentrations of THSWD,the alpha diversity of which was higher than that in the MODEL group,and beta diversity was more similar to that in the CON group.Conclusion THSWD has a good thera-peutic effect on mice with RIE,which can significantly improve gut microbial disorder and vascular function damage caused by radiation exposure.
