牡蛎多糖缓解体力疲劳及其对力竭小鼠肠道菌群的影响

目的 研究牡蛎多糖缓解体力疲劳的功能及对力竭小鼠肠道菌群结构和多样性的调节作用。方法 以总抗氧化、清除DPPH自由基、抑制超氧阴离子自由基和羟自由基能力为指标,分析抗氧化活性;采用共孵育法,以大肠埃希菌和金黄色葡萄球菌为指示菌分析抑菌活性;选取SPF级8周龄Balb/c小鼠,分别给予纯净水以及低[200 mg/(kg·mb)]、中[400 mg/(kg·mb)]、高[800 mg/(kg·mb)]剂量牡蛎多糖,持续口服干预5周,以小鼠负重游泳时间以及血乳酸、血尿素氮、肝糖原为指标测定其缓解体力疲劳功能,采集力竭小鼠盲肠内容物进行16S rDNA高通量测序,分析肠道菌群组成与多样性。结果 牡蛎多糖具有较好的抗氧化和抑菌活性,且与其浓度呈正相关。牡蛎多糖具有缓解体力疲劳功能,以高剂量效果最为显著。牡蛎多糖干预可影响肠道菌群的组成,高剂量组与对照组的beta多样性存在差异;两组间群落结构差异显著,高剂量组中Muribaculaceae的相对丰度显著增加,另枝菌属(Alistipes)、拟普雷沃菌属(Allo-prevotella)、颤螺旋菌属(Oscillibacter)相对丰度也更高;线性判别分析差异贡献证实Alistipes在高剂量组中差异显著,发挥了重要作用。结论 牡蛎多糖具有较好的抗氧化和抑菌活性,可缓解体力疲劳功能,且能调节小鼠肠道菌群结构和beta多样性,促进有益菌属而抑制有害菌属的生长,可为后续研发牡蛎多糖精准营养功效食品提供参考依据。
Effect of oyster polysaccharide in alleviating physical fatigue and on gut microbiota of exhausted mice
Objective To explore the function of oyster polysaccharide in alleviating physical fatigue,and its effect on the structure and diversity of gut microbiota in exhausted mice.Methods In vitro antioxidant activity was measured based on the total antioxidant capacity,DPPH free radical scavenging capacity,hydroxyl free radical inhibiting capacity and superoxide anion free radical inhibiting capacity.In vitro antimicrobial activity was measured through co-incubation with Escherichia coli and Staphylococcus aureus.Eight-week-old SPF Balb/c mice were separately administered purified water,and oyster polysaccharide at three different doses,800,400 and 200 mg/(kg·mb)respectively.After 5 weeks of or-al intervention,the effect of alleviating physical fatigue was measured by using the weight bearing swimming time and the levels of lactic acid,urea nitrogen and glycogen.Meanwhile,cecal contents were collected for 16S rDNA high-throughput sequencing to explore its effect on the structure and diversity of gut microbiota.Results Oyster polysaccharide exerted good in vitro antioxidant and antibacterial activities,and showed positive correlation with its concentration.It could allevi-ate physical fatigue,and the effect at high dose[800 mg/(kg·mb)]was particularly significant.It regulated the composi-tion of gut microbiota,and beta diversity in high dose group was different from that in control group;community struc-tures between the two groups were significantly different;the relative abundance of Muribagulaceae significantly in-creased,and those of Alistipes,Alloprevotella and Oscillibacter were also higher in the high dose group;Alistipes showed significant difference and played an important role in the high dose group by LEfSe.Conclusion Oyster polysaccharide shows good in vitro antioxidant and antibacterial activities,can alleviate physical fatigue,regulate the composition and beta diversity of gut microbiota,promote the growth of beneficial bacteria but inhibit harmful bacteria.This study can provide a reference basis for the subsequent development of oyster polysaccharide precise nutritional efficacy food.
