The relationship of changes in HPV L1 capsid protein,vaginal microbiota and enzymes with HR-HPV infection and CIN evolution
Objective To explore the relationship of capsid protein of human papillomavirus L1(HPV L1)and changes in vaginal microbiota and enzymes with high-risk human papillomavirus(HR-HPV)infection and the evolution of cervical intraepithelial neoplasia(CIN).Methods A total of 182 CIN patients treated in our hospital from January 2021 to March 2023 were selected,including 52 CIN grade Ⅰ patients,70 CIN grade Ⅱ patients,and 60 CIN grade Ⅲ patients.The dif-ferences in HPV L1 capsid protein,vaginal microbiota,vaginal microbial metabolites,and enzymes were analyzed and compared among the CIN patients of different grades.Results The expression of HPV L1 capsid protein in CIN grade Ⅲpatients was(20.00±5.92)PU,lower than that in grade Ⅰ and Ⅱ patients respectively(t=l2.232,10.223;all P<0.05).The expression of HPV L1 capsid protein in CIN grade Ⅱ patients was(25.10±6.04)PU,lower than that in grade Ⅰ pa-tients(t=9.922,P<0.05).The positive rate of vaginal Lactobacillus in CIN grade Ⅲ patients was 18.33%,lower than that in grade Ⅰ patients(x2=10.036,P<0.05),while the positive rate of vaginal Candida was 35.00%,higher than that in gradeⅠ patients(x2=6.892,P<0.05).The positive rates of β-Glucuronic acid(GUS),sialidase(SNA)and hydrogen peroxide(H2O2)in CIN grade Ⅲ patients were 86.67%,90.00%and 93.33%,respectively,higher than those in grade Ⅰ and Ⅱ pa-tients(x2=32.044,67.608,20.394;all P<0.05).The expression of HPV L1 capsid protein in HR-HPV infected CIN pa-tients was(24.12±3.34)PU,lower than that in uninfected patients(t=-6.150,P<0.05).The positive rates of GUS,SNA,and H2O2 in HR-HPV infected CIN patients were 65.28%,54.86%and 81.25%,respectively,higher than those in uninfec-ted patients(x2=18.637,18.440,29.397;all P<0.05).The expression of HPV L1 capsid protein in HR-HPV infected CIN grade Ⅰ patients was(28.04±3.01)PU,lower than that in uninfected CIN grade Ⅰ patients(t=-6.750,P<0.05).The pos-itive rate of H2O2 in HR-HPV infected CIN grade Ⅰ patients was 70.97%,higher than that in uninfected CIN grade Ⅰ patients(x2=4.109,P<0.05).The expression of HPV L1 capsid protein in HR-HPV infected CIN Ⅱ patients was(28.04±3.01)PU,lower than that in uninfected CIN Ⅱ patients(t=-5.165,P<0.05).The positive rates of GUS and H2O2 in HR-HPV infec-ted CIN Ⅱ patients were 50.94%and 70.97%,higher than those in uninfected CIN Ⅱ patients,respectively(x2=3.920,10.758;all P<0.05).Conclusion The expression of HPV L1 capsid protein,vaginal microbiota,metabolites and en-zymes are related to the severity of CIN,and have a certain relationship with HR-HPV infection.
