后生元那曲4580对慢性束缚应激小鼠睡眠障碍的改善效果

目的 探索后生元那曲4580对慢性束缚应激(chronic restraint stress,CRS)小鼠睡眠障碍的改善作用,为后生元制剂的功效和应用提供理论依据。方法 将225只6~8周龄、体重18~22 g SPF级C57BL/6J雄性小鼠随机分为空白对照组、CRS模型组及实验组(低剂量组、中剂量组和高剂量组),每组75只。CRS模型组和实验组连续21 d进行CRS造模,空白对照组不进行造模。实验组小鼠分别灌胃低剂量[1g/(kg·bw·d)]、中剂量[2g/(kg·bw·d)]、高剂量[3 g/(kg·bw·d)]后生元那曲4580,其余组灌胃生理盐水,灌胃持续30d。在灌胃结束后均接受糖水偏好实验、直接睡眠实验、巴比妥钠睡眠潜伏期实验、戊巴比妥钠阈下剂量催眠实验及延长戊巴比妥钠睡眠时间实验。实验结束后收集小鼠胸腺、肝脏、脾脏,称重并计算脏器系数。结果 经连续21 d造模后,与空白对照组相比,CRS模型组小鼠体重下降显著(t=6。444,P=0。001)且糖水偏好指数也显著降低(t=2。229,P=0。029)。对比CRS模型组,后生元那曲4580低剂量组小鼠体重增加显著(t=2。347,P=0。022);实验组小鼠糖水偏好指数均有所上升。在直接睡眠实验中,所有小鼠均未进入睡眠状态。在巴比妥钠睡眠潜伏期实验中,与CRS模型组相比,后生元那曲4580高剂量组的睡眠潜伏期显著增加(t=-2。575,P=0。018)。在戊巴比妥钠阈下剂量催眠实验中,后生元那曲4580中剂量组的入睡小鼠数量和入睡率显著增加(Fisher确切概率,P=0。025)。结论 后生元那曲4580不具有直接催眠的作用,但具有改善CRS小鼠的焦虑和抑郁以及睡眠障碍的作用,以中剂量改善睡眠的效果较佳。
The effect of probiotics Nagqu 4580 on sleep disorder in chronic restraint stress mice
Objective To observe the ameliorating effect of postbiotics Nagqu 4580 on sleep disorders in chronic re-straint stress(CRS)mice.Methods 225 Male C57BL/6J mice,aged 6 to 8 weeks and weighed 18-22 g,were randomly devided into control group,CRS model group and three observation groups(low-,medium-and high-dose groups)and 75 mice in each group.The modeling of CRS was carried out during continuous 21 days in CRS model group and observation groups.The mice in observation groups were gavaged with low-[1 g/(kg·bw·d)],medium-[2 g/(kg·bw·d)]and high-dose[3 g/(kg·bw·d)]of Nagqu 4580 while the other groups were gavaged normal saline,respectively,for 30 days.The sucrose preference test,direct sleep test,sodium barbital-induced sleep latency test,sub-threshold sodium pentobarbital dose-in-duced sleep test and sodium pentobarbital-induced sleep duration test were performed in all groups on Day 30 after gavage.The thymuses,livers,spleens of mice were collected and the organ coefficients were calculated after the tests.Results Compared with control group,the weight and sucrose preference index of mice in CRS model group and obser-vation groups decreased significantly after modeling of CRS(t=6.444,P=0.001;t=2.229,P=0.029).Compared with CRS model group,the weight of mice in low-dose group increased significantly(t=2.347,P=0.022)and the sucrose preference index in observation groups raised.Each group of mice didn't fall asleep in the direct sleep test.Compared with CRS mod-el group,the sleep latency in high-dose group increased significantly in the sodium barbital-induced sleep latency test(t=-2.575,P=0.018).The number and the rate of asleep mice in the sub-threshold sodium pentobarbital dose-induced sleep test increased significantly in medium-dose group(P=0.025).Conclusion Relief of axiety and depression and alleviation of sleep disorders in CRS-induced mice are the main functions of probitoics Nagqu 4580 rather than direct hypnotizing.Compared with other doses,the medium dose is better.
