Comparative study on the effects of α-1,6-glucan and oligomeric isomaltose on probiotics and human gut microbiota
Objective To compare the effects of α-1,6-glucan and oligomeric isomaltose on probiotics and human gut mi-crobiota.Methods Bacteroides fragile ferments in different media containing two types of oligosaccharides,and the growth curve was plotted by counting the live bacteria;Fresh feces from 12 healthy volunteers were collected,and the sus-pension of gut microbiota were inoculated to the control group(nitrogen-containing basic culture medium),α-1,6-glucan group(nitrogen-containing basic culture medium with 1%α-1,6-glucan),and oligosaccharide isomaltose group(nitrogen-containing base medium with 1%oligosaccharide isomaltose)at a 10%inoculation rate.The static anaerobic fermentation was carried out for 24 hours under constant temperature conditions at 37 ℃.High-throughput sequencing technology was used to analyze the microbial community structure of fecal fermentation broth;Short chain fatty acids(SCFAs)levels in fecal fermentation broth were detected by High Performance Liquid Chromatography(HPLC).Results α-1,6-glucan had a stronger ability to promote the proliferation of Bacteroides fragile compared to oligomeric isomaltose[(5.27±0.33)vs(4.80±0.66),×108 CFU/mL];Reduce the ratio of Firmicutes/Bacteroidetes in feces(t=2.821,P=0.047 8)and significantly increase the content of SCFAs(Propionate:t=3.750,P=0.009 2;Butyrate:t=5.747,P=0.001 8).Conclusion α-1,6-glucan can improve the structure of gut microbiota and promote the production of SCFAs,which may alleviate intestinal inflam-mation and alleviate obesity.
