Burkholderia pseudomallei infection case was found in Dalian by transcriptome sequencing
Objective To directly detect unknown pathogens in specimen of patient with suspected infection by using transcriptome sequencing,providing a basis for diagnosis,treatment and early warning of infection.Methods The RNA was extracted from the specimen of a patient with suspected infection in Dalian area.The RNA-Seq library based on the Ion Torrent second generation sequencing was established and sequenced.The sequencing data was analyzed using bioin-formatics to identify the specie of microorganisms present in the specimen,so as to identify the pathogens and verify them with isolation and PCR.Results The sequencing data analysis found that 2,216 sequence reads were homologous to Burkholderia pseudomallei.The verification results showed that Burkholderia mallei/Burkholderia pseudomallei was isol-ated from the specimen,and Burkholderia pseudomallei sequences were specifically amplified from the specimen without amplification of Burkholderia mallei,suggesting the presence of Burkholderia pseudomallei infection in the patient.Conclusion The melioidosis was detected in Dalian area for the first time.Transcriptome sequencing can be used to dir-ectly detect unknown pathogens in patients with suspected infection,provides a powerful technical means for early warn-ing,monitoring and handling of public health emergencies such as infectious diseases.
