Relationship between fecal short/medium chain fatty acids and constipation severity in patients with slow transmission constipation based on GC-MS method
Objective To observe the changes of fecal short/medium chain fatty acids(SCFAs/MCFAs)and their rela-tionship with the severity of constipation in patients with slow-transmission constipation(STC)using gas chromatography-mass spectrometry(GC-MS).Methods Fresh feces from 30 cases in STC group and 30 cases in normal control group in the hospital were collected,the fecal SCFAs/MCFAs were analyzed by using GC-MS,and their relationship with constipa-tion severity score(CSS)was observed.Results A total of 9 SCFAs and 4 MCFAs were detected.The total SCFAs level in the STC group was significantly lower than that in the normal control group(Z=4.355,P<0.00l),while the difference in the total MCFAs levels between the two groups was not statistically significant(Z=-1.028,P=0.304).Orthogonal partial least squares discriminant analysis(OPLS-DA)showed that there were differences in s SCFAs/MCFAs between the two groups,and this assessment model was stable(Q2=0.446,P<0.005).The results of univariate analysis showed that the levels of fecal acetic,propionic and butyric acids were down-regulated,while those of hexanoic acid and heptanoic acid were up-regulated in the STC patients compared with those in the normal control group.KEGG analysis revealed that the differential metabolites in the two groups involved 16 pathways,in which the carbohydrate digestion and absorption path-ways were significantly lower in the STC group than in the normal control group(P=0.018).Correlation analysis showed that the levels of acetic acid,propionic acid and butyric acid were negatively correlated with CSS,while heptanoic acid and hexanoic acid were positively correlated with CSS.Conclusion Fecal SCFAs/MCFAs levels in STC patients differed from those of healthy individuals,as shown by a decrease in the levels of total SCFAs,acetic acid,propionic acid and bu-tyric acid,and an increase in the levels of hexanoic acid and heptanoic acid.
