Mitigating effect of Tibetan Fritillaria cirrhosa D.Don on non-alcoholic fatty liver fibrosis in mice
Objective To explore the blocking effect of Tibetan Fritillaria cirrhosa D.Don on the fibrotic process of non-alcoholic fatty liver(NAFL)and its mechanism.Methods Thirty 6-week-old male C57BL/6 mice were randomly di-vided into control group(Group N),model group(Group M)and a Tibetan Fritillaria cirrhosa D.Don treatment group(Group H).A model of NAFL fibrosis was constructed by feeding a high-fat diet and intraperitoneal injection of carbon tet-rachloride,with the intervention of gavage of Tibetan Fritillaria cirrhosa D.Don suspension at 0.2 mL/pupil(100 mg/kg/day for each mouse).Liver lipid droplet aggregation,fibrosis status and recovery of intestinal barrier were detected by using HE staining,oil red staining and MASSON staining.Metabolic and inflammatory indexes in mice were detected with enzyme-linked immunosorbent assay and microplate assay,including fasting serum glucose,total cholesterol and tu-mor necrosis factor a.Results Compared with group M,the NAS score of liver(t=5.667,P=0.001)reduced,lipid droplet aggregation in liver and levels of AST(t=9.338,P=0.001)and ALT(t=213.6,P<0.001)markedly reduced in group H.Compared with group M,group H significantly reduced the proportion of fibers in the liver(t=2.693,P=0.036)and not-ably decreased the expression of hepatic a-SMA(t=6.295,P=0.002)and TIMP-1(t=4.494,P=0.004),and mice showed a tendency of decreased insulin resistance,and the level of serum triglyceride(t=4.253,P=0.013)significantly decreased.Compared with group M,the structures of duodenal and colonic basal lamina of model mice in group H were prominently repaired,the production of secreted mucin(t=2.989,P=0.024)observably increased,while the increase of serum LPS(t=6.690,P=0.003)was significantly inhibited.Conclusion Tibetan Fritillaria cirrhosa D.Don can significantly inhibit the process of NAFL fibrosis,while improve glucose-lipid metabolism and repair intestinal barrier.
