摘要
目的 观察EphA4基因在K562细胞野生株与耐药株中的表达情况,为研究EphA4在白血病发生发展中的作用奠定基础.方法 以荧光实时定量PCR方法检测K562细胞野生株与耐药株中EphA4基因表达.结果 E-phA4基因在K562细胞野生株中的表达明显高于K562细胞耐药株,差异有统计学意义(P<0.01).结论 根据EphA4在K562细胞野生株与K562细胞耐药株中的表达差异显著,推论EphA4这种差异的存在对于研究K562细胞所代表的慢性粒髓系白血病的作用机制、病理演变、临床表现及预后具有一定意义.
Abstract
Objective To lay the foundation for the role of EphA4 in the development of leukemia by observation of the EphA4 gene expression in wild strain and resistant strain of K562 cells. Methods EphA4 gene expression in wild strain and resistant strain of K562 cells were detected quantitatively with real-time fluorescence quantitative reverse transcription polyraerase chain reaction (QRT-PCR). Resolts The difference of EphA4 gene expression between wild type K56Z cells and resistant type K562 cells was statistically significant (P<0.01),and the EphA4 gene expression in the wild strain of K562 cells was significantly higher than that in resistant strains of K562 cells. Conclusion We make a conclusion that it is meaningful for the study of mechanism, pathological evolution,clinical manifestations and prognosis of chronic myeloid leukemia based on the significant difference of EphA4 gene expression between wild strains and resistant strains of K562 cells.
