The Protective Effect of Rabeprazole on the Gastric Mucosa in Rats
Objective To observe the protective effect of rabeprazole on the gastric mucosa in rats. Methods Gastric chamber perfusion with rabeprazolc(20 mg/kg)and intravenous injections with Nurnitro-L-arginine methyl ester(L-NAME,4 mg/kg)were administered in rats with gastric mucosal lesion induced by pure alcohol. Gastric mucosal blood flow(GMBF)was assessed with laser Doppler flowmetry(LDF),gastric mucosal and serum NO2-/N()1- ratio were measured by cadmium granulation reduction and colorimetrie method, and the changes of ulcer index and the severity of tissues necrosis and neutrophils infiltration were observed. Results The ulcer index of rats with rabeprazole treatnent was much lower than that of the control group( P<0.01) with lesser degree of tissues necrosis and neutrophils infiltration(P<0.OS). The protective effect of rabeprazole was significantly inhibited by prior administration of L-NAME that. Gastric rabeprazole administration obviously increased GMBF,gastric mucosal and serum NO2" / NO3" ratio,and such effect,apart from its action against secretion.could be counteracted by pretreatment with L-NAME. Conclusion Rabeprazole play an important protective role in the gastric mucosa in rats, which nitric oxide (NO)is involved in.
