摘要

目的 比较3种不同病毒滴度测定方法在通风系统用空气净化装置病毒净化效率试验中的适用性.方法 采用气溶胶染菌,分别使用噬斑法、定量聚合酶链反应法(qPCR)和结合细胞培养的qPCR法(ICC-qPCR)3种病毒滴度测定方法,对5款不同类型空气净化装置的一次净化效率进行对比.结果 高压静电净化装置、初效过滤器、中效过滤器的净化效率,3种方法检测结果无统计学差异(P>0.01);紫外线净化装置和高效过滤器的净化效率,qPCR法的检测灵敏性高于与其他2种方法(P<0.01),ICC-qPCR法耗时最短(约5 h),重复性好,RSD值小于其他2种方法,最低为0.01%.结论 ICC-qPCR法适用范围广、用时短、结果稳定性高,适用于通风系统用净化装置病毒净化效率的评价.

Abstract

Objective To compare three different virus titer testing methods in evaluating the virus aerosols cleaning efficiency of cleaning devices used in ventilation system.Methods After aerosol contamination,plaque assay,quantitative polymerase chain reaction(qPCR)and integrated cell culture-qPCR(ICC-qPCR)were used to detcted virus titer.One pass cleaning efficiency of 5 different types of cleaning devices were compared.Results For high voltage electrostatic purification device,primary efficiency filter and medium efficiency filter,cleaning efficiency had no significant difference among the 3 virus titer testing methods.For ultraviolet cleaning devices and high efficiency particulate air filter(P>0.01),the test sencibility of qPCR method was higher than the other two methods(P<0.01).ICC-qPCR method had the shortest time consuming(about 5 h)and had the best repeatability.RSD was lower than the other two methods,and the lowest value was<0.01%.Conclusion ICC-qPCR method has a shorter test cycle and is more stable.It is suitable for evaluating virus cleaning efficiency of the cleaning devices used in ventilation system.
