SLAF-seq-based selective genotyping and bulked segregant analysis(Super-BSA),together with bioinformatic methods were performed in three extreme phenotypes populations(sweetness,acidity and neither sweet nor sour)from the F2 population of a flavorful melon(Cucumis melo L.)‘Fengwei No. 4’;fine mapping of its genes related to sugar content and acidity traits was achieved and functional markers were developed. Results showed that:(i)both sugar content and acidity traits were mapped in parental genome;(ii)one sugar content trait-related candidate gene and three acidity trait-related genes were obtained;(iii)one sugar content(SLAF18745-S01)and three acidity(SLAF36334-S02,SLAF50072-S03 and SLAF31212-S04)trait-related gene functional molecular markers were developed based on the candidate genes. In conclu?sion,the functional molecular markers developed in this study provide an important marker foundation for molecular mark?er-assisted selection of quantitative trait loci of melons,which can reduce breeding cycles and improve the accuracy of selec?tion.
