The genome and infectious clone of papaya ringspot virus melon isolate
Papaya ringspot virus(PRSV)is one of the most important viruses in cucurbits crops.In this study,the genome sequence of melon isolate HaNHK10 was cloned and analyzed,and its full-length cDNA infectious clone was constructed.The results showed that the genome of isolate HaNHK10 was 10 332 nt in length,and the identities of the genome nucleotide sequence and amino acid sequence between HaNHK10 and other isolates were 74.60%-97.80% and 85.30%-98.50%,respectively.Phylogenetic analysis based on genome sequences showed that HaNHK10 and all the isolates from China were clustered in groupⅡ,and zucchini isolate PRSV-SD from Shandong province was most closely related to HaNHK10.The inoculation showed that the infectious clone was successfully constructed,and it could systematically infect melon,cucumber,watermelon,pumpkin,zucchini and bottle gourd.The progeny produced from the clone was infectious by mechanical inoculation.
