Exploring the mechanism of Yusi Shengjing Formula in treating asthenospermia based on network pharmacology and molecular docking technology
Objective To explore the mechanism of Yusi Shengjing Formula in treating asthenospermia.Methods The effective ingredients and target points of the core drugs of Yusi Shengjing Formula(Atractylodes macrocephala,Morinda officinalis,Lycium barbarum,Radix Rehmanniae,Radix Pseudostellariae)were screened by adapting network pharmacology research methods and using database/analysis platform of traditional Chinese medicine system pharmacological(TCMSP).The corresponding active ingredients and drug targets of deer antler gum were obtained through literature retrieval in Swiss Target Prediction.The disease targets of asthenozoospermia were obtained through Genecards and online mendelian inheritance in man(OMIM)databases,and the protein interaction network was constructed by using the search tool for recurring instances of neighbouring genes(STRING)platform for the target proteins that intersect with each other and screen for key targets.R software was used for enrichment analysis based on kyoto encyclopedia of genes and genomes(KEGG)and gene ontology(GO)databases to find the common functions and related pathways of the key genes.Protein-protein interaction(PPI)network was constructed for key genes.AutoDock was used to perform molecular docking between five Hub genes and one of the active ingredients with the highest OB value targeted.Results A total of 73 effective ingredients and 218 corresponding drug targets of the core drug of Yusi Shengjing Formula were screened out,311 drug targets were obtained for deer antler gum,3,205 asthenospermia related targets were obtained,and 230 key targets were obtained at the intersection of both.The results of functional enrichment analysis showed that key target genes were significantly correlated with responses to nutrient levels,oxygen levels,and the regulation of reactive oxygen species metabolism processes.In addition,key target genes also participated in apoptosis,hypoxia-inducible factor-1(HIF-1)signaling pathway,interleukin-17(IL-17)signaling pathway and other pathways.The results of molecular docking show that the active ingredient is well docked with the target.Conclusions The Yusi Shengjing Formula may act on key targets through quercetin and other active ingredients,which in turn participates in regulating the HIF-1 signaling pathway and IL-17 signaling pathway in asthenospermia,increases the regulation of reactive oxygen species metabolism and cellular response to oxidative stress,thereby exerting a therapeutic effect on asthenospermia.
