Expression of long-chain non-coding RNA CRNDE,micro-RNA-4262 and zinc finger E-box binding homeobox 1 in cervical cancer and their relationship with clinicopathological features and prognosis
Objective To investigate the expression of long-chain non-coding RNA(LncRNA)CRNDE,micro-RNA(miRNA)-4262 and zinc finger E-box binding homeobox 1(ZEB1)in patients with cervical cancer and their relationship with clinicopathological features and prognosis.Methods A total of 100 patients with cervical cancer admitted to Hainan West Central Hospital from June 2018 to June 2019 were selected as study subjects.The expression of LncRNA CRNDE,miR-4262 and ZEB1 mRNA in cervical cancer tissues and paracancerous tissues by real-time fluorescence quantitative polymerase chain reaction.The binding sites of LncRNA CRNDE and miR-4262 and the binding sites of miR-4262 and ZEB1 were predicted by online website,and the correlation of their expression and their relationship with clinicopathological features were analyzed.Patients with cervical cancer were divided into high expression group and low expression group according to the expression of LncRNA CRNDE,miR-4262 and ZEB1 mRNA in cervical cancer tissues.The survival curves of patients with cervical cancer with different expression of LncRNA CRNDE,miR-4262 and ZEB1 mRNA were drawn by Kaplan-Meier method.Results The expressions of LncRNA CRNDE and ZEB1 mRNA in cervical cancer tissue were higher than those in paracancerous tissue,while the expression of miR-4262 in cervical cancer tissue was lower than that in paracancerous tissue(t=13.198,25.123,11.843;P<0.01).The expression of LncRNA CRNDE was negatively correlated with the expression of miR-4262,positively correlated with the expression of ZEB1 mRNA,and the expression of miR-4262 was negatively correlated with ZEB1 mRNA(r=0.654,0.701,-0.687;P<0.01).The expressions of LncRNA CRNDE,miR-4262 and ZEB1 mRNA were correlated with international federation of gynecology and obstetrics(FIGO)staging and lymph node metastasis(P<0.05).During the follow-up of 6 to 36 months,9 cases lost follow-up and 13 cases died.the 3-year cumulative survival rate of 100 patients with cervical cancer was 86.93%.The results of Kaplan-Meier survival curve showed that the 3-year cumulative survival rate was lower in LncRNA CRNDE high expression group,miR-4262 low expression group and ZEB1 mRNA high expression group(Log-rank x2=3.899,4.132,3.891;P<0.05).Conclusions LncRNA CRNDE and ZEB1 mRNA are highly expressed and miR-4262 is lowly expressed in cervical cancer tissues.The expression changes of the three are related to FIGO stage,lymph node metastasis and prognosis.
