Analysis of the relationship between peripheral blood histone deacetylase 2,programmed death ligand 1 protein, and regulatory T-cell levels with clinicopathological features and prognosis in patients with ovarian cancer
Objective To investigate the relationship between the detection of peripheral blood histone deacetylase 2(HDAC2)and programmed death ligand 1(PD-L1)protein and regulatory T-cell(Treg)detection with clinicopathological features and prognosis in patients with ovarian cancer. Methods A total of 60 OC patients diagnosed and treated at Zhangjiakou First Hospital from July 2018 to September 2019 were selected as the study group. The patients were grouped into survival group(n=43)and death group(n=17)according to the 3-year follow-up of patients'survival condition.Another 60 patients with benign ovarian tumors diagnosed in the same hospital during the same time were selected as the control group. The levels of Treg,T helper cell 17(Th17)and Th17/Treg level in peripheral blood of all patients were detected by flow cytometry, the levels of HDAC2 and PD-L1 proteins were detected by enzyme-linked immunosorbent assay. The relationship between the levels of Treg,Th17,Th17/Treg,HDAC2 and PD-L1 proteins and the clinicopathological features was analyzed, the peripheral blood Treg,Th17,Th17/Treg,HDAC2,and PD-L1 protein levels were analyzed and compared between the survival group and the death group after follow-up for 3 years after treatment.Logistic regression was applied to analyze the factors influencing the prognosis of OC patients. Results The levels of Treg,Th17,Th17/Treg,HDAC2 and PD-L1 in patients in the study group were significantly higher than those in the control group(P<0.05).The levels of Treg,Th17,Th17/Treg,HDAC2 and PD-L1 proteins in OC patients in stage Ⅰ,stage Ⅱ,stage Ⅲ and stage Ⅳ increased successively, and the difference was statistically obvious(P<0.05).The clinical stage, residual lesion size, lymph node metastasis,Treg,Th17,Th17/Treg,HDAC2 and PD-L1 proteins levels were all influencing factors for the prognosis of OC patients(P<0.05).Conclusions Treg,Th17,Th17/Treg,HDAC2 and PD-L1 proteins participate in and affect the development of OC,which are closely related to the prognosis of patients.
