Expression of miRNA-5582 in ovarian cancer tissue and its effect on the proliferation and invasion of ovarian cancer cells
Objective To observe the effect of microRNA(miRNA)-5582 on the proliferation and invasion of ovarian cancer SK-OV-3 cells and explore its possible mechanism.Methods The University of California,Santa Cruz(UCSC)database was used to analyze miRNA-5582 expression in ovarian cancer tissue and its relationship with the clinical stage of ovarian cancer patients.Quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to analyze miRNA-5582 expression in ovarian cancer cell lines A2780,HO-8910,OC3,SK-OV-3 and normal ovarian epithelial cell lines IOSE80.The SK-OV-3 cell line with the lowest expression of miRNA-5582 were divided into control group and miRNA-5582 group,and were transfected with miR-NC mimic and miRNA-5582 mimic,respectively.MTS method and transwell assay were used to detect the proliferation and invasion of SK-OV-3 cells,respectively.The target gene of miRNA-5582 was retrieved from microRNA.org and miRWalk 3.0 databases.Dual luciferase reporter assay was used to verify the target gene of miRNA-5582.qRT-PCR and western blot were used to detect target gene expression.Results The expression of miRNA-5582 in ovarian cancer tissues was significantly lower than that in normal tissues(P<0.01).The expression level of miRNA-5582 was negatively correlated with the clinical stage of ovarian cancer patients(P<0.01).The expression of miRNA-5582 in ovarian cancer cell lines was all lower than that of IOSE80 cells(P<0.05),and the expression of miRNA-5582 in SK-OV-3 cells was the lowest(P<0.01).The relative expression of miRNA-5582 in the control group and miRNA-5582 group was 1.53±0.82 and 9.52±1.08,and the transfection system was successfully constructed(P<0.01).The cell activity of SK-OV-3 cells in the miRNA-5582 group was significantly lower than that of the control group on the third,fourth,and fifth day(P<0.05).The number of invasive cells in the control group and miRNA-5582 group were 49.48±3.78 and 15.77±3.40,respectively,and the number of invasive cells in the miRNA-5582 group was significantly less than that in the control group(P<0.01).The target gene of miRNA-5582 is serine and arginine rich splicing factor 3(SRSF3)(P<0.01).The expression of SRSF3 gene in SK-OV-3 cells in the miRNA-5582 group was significantly lower than that in the control group(P<0.01).Conclusions MiRNA-5582 is lowly expressed in ovarian cancer tissues and cell lines,and upregulated miRNA-5582 reduces the proliferation and invasion ability of ovarian cancer SK-OV-3 cells by inhibiting the expression of SRSF3 gene.MiRNA-5582 may provide a new target for the prevention and treatment of ovarian cancer.
