Analysis of expression and significance of RNF43 in uterine corpus endometrial carcinoma based on TCGA database
Objective To investigate the expression and potential role of RNF43 in uterine corpus endometrial carcinoma(UCEC).Methods RNAseq data and corresponding clinical data of UCEC tissues(n=554)and adjacent tissues(n=23)were downloaded from TCGA database,and R language was used to analyze RNF43 expression in UCEC with different clinicopathological characteristics.The correlation between prognostic value of RNF43 and prognosis of patients was analyzed by GEPIA 2.The KEGG pathway enrichment analysis of RNF43 and its co-expressed genes was performed using the online database of LinkedOmics.The correlation between RNF43 expression and immune cells in UCEC was analyzed by using the R-package"limma".Results RNF43 expression in UCEC tissue was significantly higher than that in the adjacent tissues(P<0.001).There were a statistically significant differencse in RNF43 expression among patients of different ages and tumor grades(P<0.01).Survival analysis showed that the overall survival of UCEC patients with high expression of RNF43 was longer than that with low expression of RNF43(P<0.05).Gene enrichment analysis suggested that RNF43 might be related to Wnt/β-Catenin/TCF signaling pathway,mitochondrial metabolism,lymphocyte and neutrophil mediated immunity.(FDR<0.05,P<0.05).The expression of RNF43 was positively correlated with follicular helper T cells,active memory CD4+T cells,γδT cells(R>0,P<0.05),and negatively correlated with neutrophils(R<0,P<0.05).Conclusions RNF43 can be used as a potential marker for prognosis and immunotherapy of UCEC.
