Analysis of relationship between mRNA expression levels of Toll-like receptor 2 and Toll-like receptor 4 and occurrence and outcomes of urinary tract infection after prostate cancer surgery
Objective To analyze the relationship between mRNA expression levels of Toll-like receptor 2(TLR2)and Toll-like receptor 4(TLR4)and occurrence and outcomes of urinary tract infection after radical surgery of prostate cancer(PC).Methods A total of 80 patients with urinary tract infection after PC radical surgery admitted to the Second People's Hospital of Hengshui from March 2020 to March 2023 were selected as study objects and set as infected group,and another 80 patients who did not develop urinary tract infection after PC radical surgery during the same period were selected and set as non-infected group.The mRNA expression levels of TLR2 and TLR4 and the levels of serum inflammatory factors were compared between the two groups.The correlation between mRNA expression levels of TLR2 and TLR4 and inflammatory factors was analyzed.According to the outcomes of patients in infected group after treatment,they were divided into good prognosis group and poor prognosis group,and the predictive efficiency of TLR2 mRNA and TLR4 mRNA on poor prognosis was analyzed.Results The mRNA expression levels of TLR2 and TLR4 in peripheral blood mononuclear cells and levels of serum interleukin(IL)-6,IL-8 and tumor necrosis factor-α(TNF-α)were higher in infected group compared with those in non-infected group(P<0.05).Pearson analysis showed that mRNA expression levels of TLR2 and TLR4 were positively correlated with levels of serum IL-6,IL-8 and TNF-α(P<0.05).After treatment,there were 61 patients with good prognosis in infected group(good prognosis group).Compared with poor prognosis group,the mRNA expression levels of TLR2 and TLR4 in peripheral blood mononuclear cells were higher in good prognosis group(P<0.05).The sensitivities/specificities of TLR2 mRNA,TLR4 mRNA alone and combined detection in predicting poor prognosis of urinary tract infection were 89.50%/63.90%,73.70%/73.80%and 94.70%/62.30%,and the areas under the curves(AUC)were 0.808,0.790 and 0.890 respectively.Conclusions The occurrence and outcomes of urinary tract infection after PC radical surgery are related to the mRNA expressions of TLR2 and TLR4 in peripheral blood,and their role mechanism may be related to the up-regulations of expressions of IL-6,IL-8,TNF-α and other inflammatory factors.
