The correlation between the expression of NOD-like receptor protein 3 and Toll-like receptor 4 and T helper cell 1/T helper cell 2 related cytokines in patients with syphilis serum fixation
Objective To investigate the correlation between NOD-like receptor protein 3(NLRP3)and Toll-like receptor 4(TLR4)expression and T helper cell 1/T helper cell 2(Th1/Th2)related cytokines in patients with syphilis serum fixation.Methods A total of 197 syphilis patients admitted to Santai Hospital,North Sichuan Medical College from February 2021 to April 2022 were selected as the study objects.Patients with serum negative conversion after anti syphilis treatment were selected as the negative conversion group(n=88),patients who did not receive anti syphilis treatment were collected as the syphilis group(n=45),patients with serum fixation after anti syphilis treatment were selected as the fixation group(n=64).Healthy people who underwent physical examination during the same period were selected as the control group(n=53).The relative expression levels of NLRP3,TLR4 mRNA in peripheral blood mononuclear cells(PBMCs)were detected by real-time fluorescence quantitative polymerase chain reaction.The expression level of Th1/Th2 related cytokines was detected by enzyme-linked immunosorbent assay;Pearson method was applied to analyze the correlation between NLRP3,TLR4 mRNA and Th1/Th2 related cytokines.Results The expression levels of TLR4,NLRP3 mRNA in PBMCs of all groups were compared,syphilis group>negative group>control group>fixed group,and the differences were statistically significant(P<0.05).The levels of interleukin(IL)-2 and interferon(IFN)-γ in all groups were compared,control group>negative group>syphilis group>fixed group,with statistical significance(P<0.05).The levels of IL-1β,IL-4,IL-10 and IL-18 in all groups were compared,control group<negative group<syphilis group<fixed group,with statistical significances(P<0.05).The expression of TLR4,NLRP3 mRNA were positively correlated with IFN-γ and IL-2,and negatively correlated with IL-1β,IL-4,IL-10 and IL-18 in patients with syphilis serum fixation(P<0.05).Conclusions The expression levels of NLRP3,TLR4 mRNA were significantly decreased in patients with syphilis serum fixation,and were closely related to Th1/Th2 related cytokines.
