Effect of short time insemination and overnight insemination with antagonist protocol on embryo development parameters and clinical pregnancy outcomes
Objective To explore the effects of the antagonist protocol of conventional in vitro fertilization-embryo transfer(IVF-ET)through short time insemination and overnight insemination on embryo development parameters and clinical pregnancy outcomes. Methods The retrospective analysis was performed based on the clinical data of 909 patients received IVF-ET cycles with antagonist protocol in Qinhuangdao Maternal and Child Health Care Hospital between January 2017 and June 2022.The patients were divided into short time insemination(short time group,n=424)and overnight insemination(overnight group,n=485)according to the methods of fertilization. The fertilization, embryonic development and clinical pregnancy outcome were compared between the two groups. Results The fertilization rate and normal fertilization(2PN)rate of the short time group were lower than those in the overnight group, but the polyspermy rate and blastocyst formation rate were higher than those in the overnight group, with statistically significant differences(P<0.01).There were no statistical difference of the single pronuclear(1PN)fertilization rate,non-pronuclear(0PN)fertilization rate and high quality embryo rate between the two groups(P>0.05).Comparison of the transfer cycle rate, endometrial thickness, average number of transferred embryo, implantation rate, clinical pregnancy rate, abortion rate and live birth rate between the two groups showed no statistical difference(P>0.05).Conclusions In the antagonist protocol, overnight insemination has higher 2PN rate than short time insemination, while short time insemination had more advantage in the blastocyst formation rate. They have their own advantages but do not affect the clinical outcome. For secondary infertility patients using antagonist protocol, the overnight insemination is still a kind of insemination method worthy to be affirmed.
