Objective To analyze the relationship between the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway and male infertility pregnancy outcomes of in vitro fertilization-embryo transfer/intracytoplasmic sperm injection-embryo transfer(IVF-ET/ICSI-ET),and its predictive value.Methods A total of one hundred and five male infertility patients admitted to Jinjiang District Maternal and Child Health Hospital of Chengdu from October 2022 to October 2023 were selected as the study subjects,and were divided into the successful group(n=79)and unsuccessful group(n=26)according to the IVF-ET/ICSI-ET clinical pregnancy outcomes.Baseline data,JAK2/STAT3 signaling pathway indexes were compared between the two groups,and Logistic regression model was used to analyze the IVF-ET/ICSI-ET outcomes of the patients,and the predictive value was analyzed by plotting the receiver operating characteristic(ROC)curve.Results The total number of forward-moving sperm and the percentage of normal-morphology sperm in the unsuccessful group were both lower than those in the successful group(P<0.05).The expression levels of JAK2 and STAT3 proteins in the unsuccessful group were also lower than those in the successful group(P<0.05).The total number of forward-moving sperm,the percentage of normal-morphology sperm,JAK2 and STAT3 were all protective factors for successful IVF-ET/ICSI-ET outcomes in male infertility patients(P<0.05).The ROC curve showed that the area under the curve for predicting successful IVF-ET/ICSI-ET outcomes in male infertility patients using combined predictors was larger than that of each covariate(P<0.05).Conclusions The total number of forward-moving sperm,the percentage of normal-morphology sperm,JAK2 and STAT3 are closely related to the clinical pregnancy outcomes of male infertility IVF-ET/ICSI-ET,and their combined predictive efficiency is high.
