Expression and clinical significance of cuproptosis related genes ferredoxin 1 and dihydrolipoamide transacetylase in endometrial cancer
Objective To investigate the expression and clinical significance of cuproptosis related genes such as ferredoxin 1(FDX1)and dihydrolipoamide transacetylase(DLAT)in endometrial cancer.Methods A total of 110 patients with endometrial cancer admitted to Xuzhou Central Hospital from September 2020 to August 2023 were collected as research objects,and the endometrial cancer tissue and adjacent tissues were taken during surgery.Quantitative real-time polymerase chain reaction method was applied to detect the expression levels of FDX1 and DLAT in endometrial cancer tissue and adjacent tissues,and the relationship between the expression levels of FDX1 and DLAT in the endometrial cancer tissues and the clinicopathological characteristics was analyzed.Kaplan-Meier survival curve was applied to analyze the relationship between the expression levels of FDX1 and DLAT and the 3-year prognosis of patients.Cox regression was applied to analyze the prognostic factors of endometrial cancer patients.Results The expression level of FDX1 in the endometrial cancer tissue was lower than that in the adjacent tissue,while the expression level of DLAT was higher than that in the adjacent tissue(P<0.05).The proportions of low FDX1 expression and high DLAT expression in patients with international federation of gynecology and obstetrics(FIGO)stages Ⅲ-Ⅳ,lymph node metastasis,myometrial infiltration ≥1/2,and vascular infiltration were higher than those in patients with FIGO stages Ⅰ-Ⅱ,no lymph node metastasis,myometrial infiltration<1/2,and no vascular infiltration(P<0.05).The 3-year survival rate of patients with low FDX1 expression was lower than that of patients with high FDX1 expression(P<0.05).The 3-year survival rate of patients with high DLAT expression in endometrial cancer tissue was lower than that of patients with low DLAT expression(P<0.05).FIGO staging and DLAT were the independent risk factors for 3-year mortality in endometrial cancer patients,while FDX1 was a protective factor for 3-year mortality(P<0.05).Conclusions FDX1 is low expression and DLAT is high expression in endometrial cancer tissue,these two are related to the clinicopathological characteristics and prognosis of endometrial cancer patients.They may become the new action targets for the diagnosis,treatment,and prognosis improvement of endometrial cancer.
