Study on Detection Method for LAMP Technique in Clostridium Botulinum of Pet Food
The detection method for LAMP technique in Clostridium botulinum of pet food was studied in the paper.A set of primers was designed according to the specific gene(NtnH)fragments of Clostridium botulinum.Through ampilifying gene fragments of Clostridium botulinum and 14 nonClostridium botulinum,screening test of optimum temperature and response time on LAMP reaction system,specific test by using nephelometry and staining method,sensitivity test of that compared to PCR were studied.The result demonstrated that LAMP only amplified gene of Clostridium botulinum,and had a good specificity.The optimum response temperature of the system was 64℃,and the optimum response time was 60 min.The minimum detectable concentration was 0.0001 pg/μL,which was 10 times higher than PCR.The method established by the study had a good specificity and high sensitivity and short response time,which was adapt to the detection for Clostridium botulinum of pet food.
