猪卵巢circ_mmp16的鉴定及功能初探

A Preliminary Study on the Identification and Function of Circ_mmp16 in Porcine Ovaries

猪卵泡颗粒细胞的增殖能力与繁殖性状密切相关,本研究旨在鉴定猪卵巢组织的circ_mmp16 及其调控网络,并对其潜在的调控功能进行分析.运用miRanda、RNA hybird等软件对circ_mmp16 靶向调控的miRNA及靶基因进行生物信息学分析.结果显示,circ_mmp16 是存在于哺乳动物中并且物种间保守的环状RNA,由基质金属蛋白酶 16(Matrix Metallopeptidase 16,Mmp16)的 2~4 号外显子组成.此外,circ_mmp16 可作为ssc-miR-217、ssc-miR-124a、ssc-miR-16 等 26 个miRNA的竞争性内源RNA调控下游靶基因.通过对circ_mmp16-miRNA调控网络的靶基因进行GO和KEGG分析发现,Wnt信号通路、钙离子的转运、谷胱甘肽代谢以及花生四烯酸的代谢都与卵巢功能相关.组织表达谱分析发现circ_mmp16 在骨骼肌中表达量最高,在卵巢中的表达量仅次于骨骼肌.在猪卵巢颗粒细胞水平上发现circ_mmp16 具有促进细胞周期标志基因表达的作用,并能促进GPX3、GPX4 谷胱甘肽合成标志基因的表达.综上,circ_mmp16 作为调节多个miRNA的竞争性内源RNA,可能通过促进卵巢颗粒细胞的增殖以及抗氧化应激的能力起到保护卵巢颗粒细胞的作用,为进一步揭示颗粒细胞参与调控猪卵泡发育和闭锁提供基础数据.
The proliferative capacity of porcine follicular granulosa cells is closely related to reproductive traits.The aim of this study was to characterize circ_mmp16 and its regulatory network in porcine ovarian tissues,and to analyze its potential regulatory functions.The miRNAs and target genes targeted for regulation by circ_mmp16 were bioinformatically analyzed using miRanda and RNA hybird.The results showed that circ_mmp16 is a cyclic RNA present in mammals and conserved among species and consists of exons 2~4 of Matrix Metallopeptidase 16(Mmp16).In addition,circ_mmp16 can act as a competitive endogenous RNA for 26 miRNAs such as ssc-miR-217,ssc-miR-124a,and ssc-miR-16 to regulate downstream target genes.GO and KEGG analysis of the target genes of the circ_mmp16-miRNA regulatory network revealed that the Wnt signaling pathway,calcium transport,glutathione metabolism,and arachidonic acid metabolism were associated with ovarian function.Tissue expression profiling revealed that circ_mmp16 was most highly expressed in skeletal muscle,and second only to skeletal muscle in the ovary.At the level of porcine ovarian granulosa cells,circ_mmp16 was found to have a role in promoting the expression of cell cycle marker genes and to promote the expression of GPX3 and GPX4 glutathione synthesis marker genes.In conclusion,circ_mmp16,as a competitive endogenous RNA regulating multiple miRNAs,may play a role in protecting ovarian granulosa cells by promoting their proliferation as well as their resistance to oxidative stress,providing basic data to further reveal the involvement of granulosa cells in the regulation of follicular development and atresia in pigs.
