摘要

目的 分析卵巢癌(OC)中HOXB2中的表达,探讨HOXB2在OC中的潜在诊断和预后价值.方法 从Gene Expression Omnibus(GEO)数据库获得OC的基因数据(GSE66957).然后,利用STRING数据库和Cytohubba软件筛选差异表达的基因(DEGs),筛选出核心基因HOXB2,并用基因组百科全书(KEGG)和基因本体论(GO)途径评估了DEGs基因参与OC的关键途径.通过人类蛋白质图谱(HPA)数据库分析OC中HOXB2的表达.该研究纳入88 例OC患者,收集患者相关信息,使用Kaplan-Meier评估HOXB2表达量和临床特征对OC患者生存率的影响.通过实时荧光定量PCR(RT-qPCR)和免疫组化检测OC组织及其癌旁组织中HOXB2的表达水平.结果 该研究中,从数据集中获得了5 003 个差异基因,其中包括2 733 个上调基因和2 270 个下调基因,其中HOXB2是显著上调基因.KEGG分析表明,常见的DEGs与细胞黏附分子和金黄色葡萄球菌有关.GO分析表明,常见的DEGs主要参与上皮管形态发生、含有胶原的细胞外基质的调控.基于HPA数据库,HOXB2在OC中高表达.Kaplan-Meier分析表明,HOXB2的高表达与较差的总生存率显著相关.qPCR结果也证明了这一点,与癌旁相比,OC组织中HOXB2表达显著升高;免疫组化结果显示HOXB2在OC组织中明显高表达,差异有统计学意义.结论 该发现有助于进一步探索OC的发展机制,并为未来OC治疗的一个有前途的选择.

Abstract

Objective To analyze the expression of HOXB2 in ovarian cancer(OC)and explore its potential diagnostic and prognostic value in OC.Methods The gene data of OC(GSE66957)were obtained from the Gene Expression Omnibus(GEO)database.Then,using the STRING database and Cytohubba software,differentially expressed genes(DEGs)were screened,and the core gene HOXB2 was identified.The key pathways of DEGs involved in OC were evaluated using the Genomic Encyclopedia(KEGG)and Gene Ontology(GO)pathways to analyze the expression of HOXB2 in OC through the Human Protein Atlas(HPA)database.In addition,this study included 88 OC patients,collected patient related information,and used Kaplan-Meier to evaluate the impact of HOXB2 expression and clinical features on the survival rate of OC patients.The expression level of HOXB2 in OC tissue and its adjacent tissues was detected by real-time fluorescence quantification(RT-qPCR)and immunohistochemistry.Results 5 003 DEGs were obtained from the dataset,including 2 733 upregulated genes and 2 270 downregulated genes,with HOXB2 being a significantly upregulated gene.KEGG analysis indicated that common DEGs were associated with cell adhesion molecules and Staphylococcus aureus.GO analysis showed that common DEGs were mainly involved in epithelial tube morphogenesis and regulation of extracellular matrix containing collagen.Based on the HPA database,HOXB2 was highly expressed in OC.Kaplan Meier analysis showed a significant correlation between high expression of HOXB2 and poorer overall survival.The qPCR results also confirmed this,as compared to the adjacent tissues,the expression of HOXB2 in OC tissues was significantly increased.The immunohistochemical results showed that HOXB2 was significantly overexpressed in ovarian cancer tissue,with statistical significance.Conclusion These findings help to further explore the development mechanism of OC and provide a promising option for future treatment of OC.
