摘要
目的 探讨核糖核酸聚合酶Ⅱ相关蛋白3(RPAP3)在卵巢癌细胞中所发挥的生物学功能,深入探究RPAP3对卵巢癌进展的调控作用.方法 使用GENT2数据库分析RPAP3在卵巢癌中的表达,通过RT-qPCR和Western blot检测正常卵巢上皮细胞系(IOSE80)和卵巢癌细胞系(OVCAR3和SKOV3)中RPAP3 mRNA和蛋白的表达.使用慢病毒sh-RPAP3感染OVCAR3细胞系构建RPAP3敲低稳转株,采用CCK-8、伤口划痕实验和Transwell小室侵袭实验检测敲低RPAP3后在体外对卵巢癌细胞增殖、迁移和侵袭功能的影响;通过流式细胞术检测敲低RPAP3对卵巢癌细胞凋亡情况的影响.结果 从GENT2数据库中得知RPAP3在卵巢癌高表达,在卵巢癌细胞系OVCAR3及SKOV3中RPAP3 mRNA和蛋白表达水平高于正常卵巢上皮细胞系IOSE80.敲低RPAP3抑制卵巢癌细胞的增殖、迁移和侵袭,敲低RPAP3增加卵巢癌细胞凋亡率.结论 RPAP3在卵巢癌中高表达,促进卵巢癌细胞增殖、迁移和侵袭;抑制RPAP3诱导卵巢癌细胞凋亡,为卵巢癌靶向治疗提供新的潜在靶点.
Abstract
Objective To gain insight into the biological functions played by RNA polymerase Ⅱ-associated protein 3(RPAP3)in ovarian cancer cells and to conduct a comprehensive investigation into the regulatory role of RPAP3 on ovarian cancer progression.Methods The expression of RPAP3 in ovarian cancer was analyzed using the GENT2 database.RT-qPCR and Western blot were employed to detect RPAP3 mRNA and protein in normal ovarian epithelial cell lines(IOSE80)and ovarian cancer cell lines(OVCAR3 and SKOV3).Lentiviral sh-RPAP3 was used to infect OVCAR3 cell lines to construct RPAP3 knockdown stable transfections.The effects of RPAP3 knockdown on the proliferation,migration,and invasion functions of ovarian cancer cells were examined using CCK-8,wound scratch assay,and Transwell invasion assay.Additionally,the effects of RPAP3 knockdown on ovarian cancer cells were examined using flow cytometry.Results The GENT2 database indicated that RPAP3 was highly expressed in ovarian cancer.Furthermore,the mRNA and protein expression levels of RPAP3 in ovarian cancer cell lines OVCAR3 and SKOV3 were higher than those of the normal ovarian epithelial cell line IOSE80.Knockdown of RPAP3 inhibited the proliferation,migration,and invasion of ovarian cancer cells,and knockdown of RPAP3 increased the apoptosis rate of ovarian cancer cells.Conclusion RPAP3 is highly expressed in ovarian cancer and promotes ovarian cancer cell proliferation,migration,and invasion.Inhibition of RPAP3 induces apoptosis in ovarian cancer cells,and it may be a potential target for targeted therapy of ovarian cancer.
