Functional Study on the Role of NtJAZ9 Gene in Regulating the Formation of Tobacco Glandular Trichomes
[Purpose]This study aims to explore the molecular mechanism of jasmonic acid in regulating tobacco trichomes glandular trichomes occurrence.[Methods]Homologous cloning,bioinformatics analysis and gene function studies of NtJAZ9,a key repressor of jasmonic acid signaling,were conducted[Results](1)NtJAZ9 has two homologous sequences,NtJAZ9-1 and NtJAZ9-2,with coding regions of 1008 bp and 996 bp,respectively.(2)Subcellular localization results indicate that the NtJAZ9 protein is localized in the nucleus.In various tissues,the relative expression of NtJAZ9-1 is higher than that of NtJAZ9-2,with the highest expression of NtJAZ9-1 in stem epidermis and the highest expression of NtJAZ9-2 in leaves.(3)Overexpression of NtJAZ9 results in reduced glandular trichome density on leaves,while knockout of the NtJAZ9 gene significantly increases glandular trichome density;the difference in protective trichome density is not significant.Methyl jasmonate(MeJA)can significantly promote the occurrence of long-stalk glandular trichomes.After spraying,the density of long-stalk glandular trichomes increased by 1.76 times in overexpressed strains and by 0.36 times in knockout strains.[Conclusion]The NtJAZ9 gene is involved in the regulation of occurrence of tobacco glandular trichomes and affects the regulatory role of MeJA signals on the formation of tobacco glandular trichomes to a certain extent.
