Objective Explore free fatty acid(FFAs) to proximal convoluted tubules epithelial cells(HK-2) influence and function mechanism. Methods People proximal convoluted tubules epithelial cells(HK-2), were cultured with DMEM-F12 of different concentrations of palmitic acid(0, 125, 250. 500μmol) (using defat bovine serum albumin as vector), normal medium as control grop. on the 0 hour, 24 hour, 48 hour respectively: Applyied four methyl azo thiazole blue(determined by MTT) colorimetric detection different environment HK-2 cell proliferation capacity;Streaming cells in cell apoptosis FFAs intraoperative detection and the influence of the cell cycle; Immune cell chemical method before and after treatment were detected in HK-2 cells interleukin-β(IL-β), interleukin-8 (IL-8) expression changes. Results In FFAs doses and time dependence inhibit HK-2 cell proliferation(P<0.05), and possibly by blocking the cell cycle and apoptosis(P<0.05). After FFAs role by IL-β, IL-8 expression increase. Conclusion Free fatty acid HK-2 it inhibits cell proliferation, promote its apoptosis role, and inflammatory cells factor IL-β, IL-8 express increasing, participated in its occurrence and development process.
