N乙酰化转移酶2基因多态性与异烟肼所致肝损伤研究进展

N-乙酰转移酶2是人体内重要的Ⅱ相代谢酶,在芳香胺类和肼类化学物质代谢中起重要作用。我国人群中NAT2基因以快乙酰化基因表型为主,最常见的基因型为NAT2*4A、NAT2*6A、NAT2*7B。NAT2慢乙酰化代谢基因型与异烟肼所致的药物性肝炎相关,NAT2*5等位基因频率的差异是导致慢乙酰化频率差异的主要原因之一。研究NAT2基因多态性对指导异烟肼个体化治疗、减少或减轻药物不良反应有着重要的意义。
N-acetyl transferase 2 is an important Ⅱ metabolic enzymes in human bodies, it plays an important role in metabolism of aromatic amine and the hydrazine. NAT2 gene is given priority to fast acetylated gene phenotypes in Our country, the most common genotype are NAT2*4A, NAT2*6A, NAT2*7B. The Slow NAT2 genotypes with isoniazid acetylated metabolism is related to the drug-induced hepatitis, differences of NAT2*5 in allele frequency is one of the main reasons that cause slow frequency of acetylated for the differences. Research of NAT2 gene polymorphisms has important significance in isoniazid individualized treatment and reducing or mitigating adverse drug reaction.
