IL-2基因敲除小鼠的饲养繁育及基因型鉴定

目的繁殖和鉴定白介素2(IL-2)基因敲除小鼠。方法将所引进的杂合子小鼠进行饲养并繁殖,提取子代小鼠尾部DNA,用PCR方法检测子代小鼠基因型。结果子代小鼠基因型分别为杂合子(IL-2+/-)、纯合子(IL-2-/-)和野生型(IL-2+/+)。结论 IL-2基因敲除小鼠的繁育和鉴定均获得成功,为进一步研究IL-2缺失如何影响机体免疫功能提供了必要条件。
Breeding reproducing and genotype identifying IL-2 knockout mice
Objective To breed, reproduce and identify IL-2 knockout mice. Methods Heterozygote mice were bred and reproduced. Genome DNA extracted from the murine tail was subjected to PCR fog genotype identification. Result The genotypes of the offspring mice were heterozygous(IL-2+/-), homozygous(IL-2-/-), and wild-type(IL-2+/+). Conclusion Breeding and reproducing of IL-2 knockout mice were successful, which provide a necessary conditions for the further study of IL-2 deletion affect immune function.
