Objective To investigate whether Cyclosporin A(CsA) affect the expression of oligodendrocyte-myelin glycoprotein(OMgp) after acute spinal cord injury in rats. Methods SD rats were divided into three groups: the control group(group A), the spinal cord injury group(group B) and the spinal cord injury with CsA -treated group(group C). The rats of group C were administered CsA through the tail vein at the early stage of spinal cord injury. The movements of hind limbs were observed in all groups and evaluated by BBB score, after that the rats were sacrificed and the injured spinal cord was taken out. The structural change of the rat spinal cord was evaluated by HE staining. The expression of OMgp protein and OMgpmRNA in the rat spinal cord and neighbor areas were detected by immunohistochemistry staining and Real-time Fluorescence Quantitative PCR(RT-PCR), respectively. Results The functional recovery of hind-limb was observed in group B and C at each study time point. After spinal cord injury, the expressions of OMgp at each study time point in group B and C were significantly higher than that of group A(P<0.05), and reach its apex at day 7. Meanwhile, the expression of OMgp at each study time point in group C was significant lower than that of group B(P<0.05). Conclusion The expression of OMgp significantly increased after spinal cord injury, and early CsA administration strongly inhibit the expression of OMgp.
