In vivo study on the reversal of cisplatin resistance of esophageal cancer by Lentiviral vector of siRNA targeting to GST-π, polβ
Objective To investigate the reversal of cisplatin resistance of esophageal cancer by Lentiviral vector of siRNA targeting to GST-π, polβ in vivo. Methods EC9706 and EC9706/cDDP cells were subcutaneously injected into athymic nude mice to set up the animal models of grafted tumor. Injecting cDDP plus GSTsi2(polsi1) to xenografts derived from drug resistant cells to detect xenografts cells changes in drug sensitivity. Results The tumor volume of the group of cDDP plus GSTsi2 was lower than that of groups of PBS and cDDP; while the tumor volume of the group of cDDP plus polsi1 had no significant difference with groups of PBS and cDDP. Conclusion Injecting cDDP plus GSTsi2 to xenografts derived from drug resistant cells could inhibit its growth; while injecting cDDP plus polsi1 to xenografts could not inhibit its growth.
