摘要
目的探讨血小板糖蛋白HPA-2基因型在血小板捐献者中的分布频率。方法收集血小板捐献者184人，其中A型44人，B型44人，O型55人，AB型41人，应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)扩增GP HPA-2基因，产物经限制性内切酶Hin1 I消化后确定其基因型。结果在四种不同血型血小板捐献者中，HPA-22a/2a基因型频率分别是0.95、0.91、0.85、0.98；2a/2b分别是0.05、0.07、0.15、0.02；2b/2b只在B型捐献者中发现1例，基因型频率是0.02。结论HPA-2基因型频率在四种不同血型血小板捐献者中的分布无显著差异。
Abstract
Objective To evaluate the distributive characteristic of genotype of platelet glycoprotein HPA-2 in the volunteer platelet donors. Methods 184 volunteer platelet donors at mobile place were collected, including 44 type A, 44 type B, 55 type O and 41 type AB volunteer platelet donors. HPA-2 polymorphism was tested by polymerase chain reaction-restricted fragment length polymorphism(PCR-RFLP) amplification. Genotyping for HPA-2 was performed by restriction enzyme analysis. Results HPA-2 2a/2a genotype frequency was 0.95, 0.91, 0.85, 0.98 respectively; 2a/2b genotype frequency was 0.05, 0.07, 0.15, 0.02 respectively; 2b/2b genotype was only one and genotype frequency was 0.02 among the 184 volunteer platelet donors with different blood type. Conclusions There is no significant difference of HPA-2 genotype frequency among the volunteer platelet donors of four different blood type.
