Effects of Schisandrin B on Lung mRNA Expression of MMP-9 and TIMP-1 in Rat Lungs Exposed to Silica
Objective To investigate the effects of schisandrin B(Sch-B) on expression of MMP-9 and TIMP-1 in rat lungs exposed to silica, and explore the intervention mechanism of Sch-B on pulmonary fibrosis induced by SiO2. Methods 48 Wistar rats were randomly divided into control group, silica group and Sch-B treated group with 16 rats in each group. The rats were explored to SiO2 by direct tracheal instillation to establishe the Silicotic animal models. Silica group and Sch-B treated group were treated with 1ml(50mg/ml) for each rat. From the first day of model establishment, Sch-B treated group were orally given(80mg·kg-1·d-1) a day, control group and SiO2 group were orally given olive oil. On the 7th and 28th days after establishment. eight rats in each group were sacrificed and samples were collected. The expressions of MMP-9 and TIMP-1 mRNA in the lung tissue were detected by RT-PCR. Results The expressions level of MMP-9 mRNA in silica group were significantly higher than those in the control groups(P<0.01 or P<0.05), but the expressions level of TIMP-1 mRNA in silica group were significantly higher than those in the control group on 7th days(P<0.05). The expressions level of MMP-9 mRNA in Sch-B treated group were significantly lower than those in the silica group on 7th days(P<0.05), but there were no significant differences of the TIMP-1 mRNA expressions between silica group and Sch-B treated group(P>0.05). Conclusion The suppression mechanism of Sch-B on pulmonary fibrosis induced by SiO2 may be related to its effects of decreasing the expressions of MMP-9 mRNA at the early stage and each period of time adjusting the expressions of MMP-9 and TIMP-1 to some extent ,so that the MMP-9/TIMP-1 tends to balance.
