兔瘟2型VP60基因生物信息学分析

2020年4月,兔瘟2型病毒病例首次在四川省被检测到,该毒株迅速在我国蔓延.VP60基因是RHDV2编码病毒衣壳蛋白主要结构蛋白,参与病毒的复制、装配和释放等过程.因此通过兔场分离RHDV2,对VP60基因测序后进行生物信息学分析,FJ fenli RHDV2-VP60基因与RHDV2-VP60参考序列的同源性为95.7%~99.4%,与RHDV-VP60参考序列的同源性为81.6%~82.5%之间,VP60蛋白主要以疏水性氨基酸为主,二级结构α-螺旋占比为8.98%,β-折叠占30.22%,β-转角占6.22%,无规则卷曲结构占54.58%,有3个跨膜片段和30个抗原位点,最大抗原位点分值为1.213.证明其研究对了解RHDV2生物学特性和致病性机制具有重要意义,为该病防控提供前期理论基础.
Bioinformatics Analysis of Rabbit Hemorrhagic Disease Virus Type 2 VP60 gene
In April 2020,the first case of rabbit distemper type 2 virus detected in Sichuan Province,the strain quickly spread in China.The VP60 gene is the main structural protein encoding the viral capsid protein of RH-DV2,involved in the rep-lication,assembly,and release processes of the virus.Therefore,this study isolated RH-DV2 from a rabbit farm and sequenced the VP60 gene for bioinformatics analysis.The homology between the FJ fenli RHDV2-VP60 gene and the RHDV2-VP60 reference sequence ranged from 95.7%to 99.4%,and the ho-mology with the RHDV-VP60 reference sequence ranged from 81.6%to 82.5%.The VP60 protein was mainly composed of hydrophobic amino acids,with a secondary structure of α-The proportion of spirals is 8.98%,β-Folding accounts for 30.22%,β-Corners account for 6.22%,irregular curly structures account for 54.58%,with 3 transmem-brane segments and 30 antigen sites,the maximum antigen site score is 1.213.This study is of great significance for understanding the biological characteristics and pathogenic mechanism of RHDV2,and provides a preliminary theoretical basis for the prevention and control of this disease.
