摘要

目的 探讨黄芪在肿瘤的化学治疗中的最佳给药时间点.方法 按照实验方案,分组、造模、给药、称重,测量肿瘤大小,并持续观察、记录小鼠的生活情况;观察肿瘤细胞的侵袭情况,计算器官指数,检测淋巴细胞数和中性粒细胞数.结果 与M组比较,5-氟尿嘧啶(5-FU)组小鼠的生活质量无明显变化,瘤体明显减小(P<0.05);与5-FU组比较,黄芪可明显改善小鼠的生活状况,进一步抑制肿瘤的生长,5-FU后+Hm组小鼠的瘤体减小具有统计学意义(P<0.05);M组肿瘤明显破坏周围组织,周围组织中新生血管丰富;与 M 组比较,5-FU 组、H 组的肿瘤细胞对周围正常组织的侵袭程度依次逐渐减弱,新生血管依次逐渐减少,且H组中形成了明显的包膜、新生血管少且管径小;与M组比较,5-FU组胸腺指数和脾指数明显下降(P<0.05);与 5-FU组比较,H组胸腺指数和脾指数明显升高(P<0.05);与M组比较,5-FU组淋巴细胞显著降低(P<0.05)、中性粒细胞显著升高(P<0.05);与5-FU组比较,H组的淋巴细胞显著升高(P<0.05)、中性粒细胞明显降低(P<0.05).结论 黄芪可增强 5-FU的抗肿瘤作用,改善 5-FU导致的生命质量低下状态,且化疗后给予黄芪效果更优.

Abstract

Objective To explore the optimal time of astragalus,s anti-tumor in chemotherapy of tumor.Methods According to the experimental scheme,Kunming mices were randomly divided into different groups,then were molded,administered and weighed,and tumor volume were measured.At the same time,we observed and recorded their living conditions during administering.The invasion of tumor cells were observed by HE staining,thymus and spleen were sacrificed to calculated thymus index and spleen index,the number of lymphocyte and neutrophil in the blood mice were detected by an automatic blood analyzer.Results Compared with the M group,the life quality of 5-fluorouracil(5-FU)group had no significant change,and the tumor size was significantly reduced(P<0.05).Compared with the 5-FU group,Astragalus could significantly improve the living conditions of mice and further inhibit the growth of tumors,the tumor size of mice in the 5-FU +Hm group decreased significantly(P<0.05).The tumor in group M obviously destroyed the surrounding tissue,and the surrounding tissue was rich in new blood vessels.Compared with group M,the invasion degree of tumor cells in group 5-FU and group H to the surrounding normal tissue was gradually weakened,and the new blood vessels were gradually reduced,and the obvious envelope,fewer new blood vessels and small tube diameter were formed in group H.Compared with group M,thymus index and spleen index in group 5-FU were significantly decreased(P<0.05).Compared with 5-FU group,thymus index and spleen index in H group were significantly increased(P<0.05).Compared with group M,the lymphocytes in group 5-FU were significantly decreased(P<0.05)and neutrophils were significantly increased(P<0.05).Compared with 5-FU group,the lymphocytes in H group were significantly increased(P<0.05)and neutrophils were significantly decreased(P<0.05).Conclusion Astragalus could enhance the anti-tumor effect of 5FU and improve the quality of life caused by 5-FU.The effect of Astragalus after chemotherapy was better than that of Astragalus when given at the same time.
