额尔敦-乌日勒对β-淀粉样蛋白25~35所致阿尔茨海默病模型大鼠脑内神经元凋亡的影响

摘要
目的 探讨额尔敦-乌日勒对β-淀粉样蛋白(Aβ)25~35所致阿尔茨海默病模型大鼠脑内神经元凋亡的影响.方法 将Aβ25~35 所致阿尔茨海默病模型大鼠(n=54)随机分为3 组:模型组、低剂量组、高剂量组.3 组分别给予等体积的0.9%氯化钠注射液、50 μg/ml额尔敦-乌日勒、100 μg/ml额尔敦-乌日勒灌胃,1 次/d,连续灌胃14 d.比较 3 组大鼠治疗不同时间点水迷宫逃避潜伏期,血清超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平,Caspase3 蛋白表达水平.结果 治疗7 d、14 d,低剂量组、高剂量组大鼠水迷宫逃避潜伏期短于模型组(P<0.05),且高剂量组短于低剂量组(P<0.05);治疗7 d、14 d,低剂量组、高剂量组大鼠血清SOD水平高于模型组(P<0.05),血清MDA水平低于模型组(P<0.05),且高剂量组SOD、MDA水平变化幅度优于低剂量组(P<0.05);治疗7 d、14 d,低剂量组、高剂量组大鼠血清TNF-α、IL-6水平低于模型组(P<0.05),且高剂量组低于低剂量组(P<0.05);治疗7 d、14 d,低剂量组、高剂量组神经元组织的Caspase3 蛋白相对表达水平低于模型组(P<0.05),且高剂量组低于低剂量组(P<0.05).结论 额尔敦-乌日勒在Aβ25~35 所致阿尔茨海默病模型大鼠的应用能缩短水迷宫逃避潜伏期,提高血清SOD水平,降低血清TNF-α、IL-6、MDA水平,还可抑制神经元细胞凋亡.
Abstract
Objective To explore the effect of Erden-Ujile on neuronal apoptosis induced by β-amyloid protein(Aβ)25~35 in Alzheimer's disease model rats.Methods The model rats of Alzheimer's disease induced by Aβ25~35(n=54)were randomly divided into three groups:model group,low dose group and high dose group.Three groups were given equal volume of saline,50 μg/ml Erdun-Uzhle and 100 μg/ml Erden-Ujile gavage once a day for 14 days,respectively.The levels of serum superoxide dismutase(SOD),malondialdehyde(MDA),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and the expression of Caspase3 protein were compared in 3 groups of rats treated with water maze at different time.Results After 7 and 14 days of treatment,the water maze escape latency of rats in low-dose and high-dose groups was shorter than that in model group(P<0.05),and that in high-dose group was shorter than that in low-dose group(P<0.05).After 7 and 14 days of treatment,the serum SOD level in low-dose and high-dose groups was higher than that in model group(P<0.05),and the serum MDA level was lower than that in model group(P<0.05),and the change range of SOD and MDA levels in high-dose group was better than that in low-dose group(P<0.05).After 7 and 14 days of treatment,the serum levels of TNF-α and IL-6 in low-dose and high-dose groups were lower than those in model group(P<0.05),and those in high-dose group were lower than those in low-dose group(P<0.05);After 7 and 14 days of treatment,the relative expression level of Caspase3 protein in neurons of low-dose and high-dose groups was lower than that of model group(P<0.05),and that of high-dose group was lower than that of low-dose group(P<0.05).Conclusion The application of Erden-Uhelle in Aβ25~35 induced Alzheimer's disease model rats can shorten the water maze escape latency,increase serum SOD level,decrease serum TNF-α,IL-6,MDA levels,and inhibit neuronal cell apoptosis.
