Optimization of Extraction and Inclusion Process of Paeonol from Moutan Peel by Plackett Burman Design
Objective To optimize the extraction and cladding process of koaypanol in peony peel by Plackett-Burman experimental design.Methods Taking the extraction rate of paeonol as an index,the extraction and encapsulation process was optimized by the star point design-effect surface method,and the extraction process of peony peel was optimized by the design expert design based on the investigation factors of temperature immersion time,water addition and distillation time for the extraction of paeonol.Using the saturated aqueous solution method,taking the encapsulation rate of koayptol as the main assessment index,taking the molar ratio of β-cyclodextrin and paeonol,stirring time and wrapping temperature as the investigation factors of paeonol adhesion,the design expert was used to design and optimize the β-cyclodextrin cladding process of paeonol.Results Using the response surface analysis of Design expert 13.0 software,it was determined that the optimal extraction process was 1.68 h immersion time,13.41 times water addition,and distillation time 3.28 h,under which the theoretical value of paeonol extraction predicted by the regression model could reach 78.81%.The optimal cladding process of paeonol was determined to be a molar ratio of 0.91:1 between β-cyclodextrin and paeonpo,a cladding time of 3.12 h,and a cladding temperature of 48.6 h,under which the theoretical value of the cladding rate of dipamarine carboxyl predicted by the regression model could reach 59.34%.Conclusion The mathematical model constructed by the star point design-effect surface method has good predictive ability,and the extraction rate and encapsulation rate of paeonol in peony peel optimized by this method can be significantly improved.
