稽延期海洛因依赖者与健康人群肠道菌群的差异分析

摘要
目的 比较稽延期海洛因依赖者与健康人群肠道菌群的差异,探讨海洛因依赖者的肠道菌群与稽延性戒断症状的关系.方法 选择宁夏回族自治区内各强制隔离戒毒所的稽延期海洛因依赖者及招募的健康人群作为研究对象,并分成稽延期组(n=36)和健康组(n=36).采用高通量16SrDNA测序技术检测两组的肠道微生物群落结构.结果 稽延期组肠道内的OTU数目高于正常组;两组肠道菌群的Alpha多样性差异无统计学意义(P>0.05);两组肠道菌群的Beta多样性差异具有统计学意义(P<0.05);在门水平上,厚壁菌门、放线菌门和变形菌门是两组的优势菌门,但丰度不同,稽延期组肠道拟杆菌门、脱硫杆菌门的相对丰度均高于正常组(P<0.05);在属水平上,与正常组相比,稽延期组肠道中拟杆菌属相对丰富(P<0.05),而纺锤链杆属的相对丰度低(P<0.05).LEfSe分析显示,稽延期组肠道菌群在门/纲/目上,拟杆菌的丰度在菌群差异中起主要作用.结论 稽延期海洛因依赖者与健康人群的肠道微生物群落结构存在差异,原因可能与稽延性戒断症状影响海洛因依赖者的肠道菌群结构相关.
Abstract
Objective To compare the difference of intestinal flora between heroin addicts and healthy people,and to explore the relationship between intestinal flora and prolonged withdrawal symptoms in heroin addicts.Methods Heroin addicts with extended period and healthy people recruited from compulsory isolation drug rehabilitation centers in Ningxia Province were selected as the study subjects,and divided into delay period group(n=36)and healthy group(n=36).High-throughput 16SrDNA sequencing technology was used to detect the intestinal microbial community structure of the two groups.Results The number of OTU in intestinal tract of delay period group was higher than that of normal group.There was no significant difference in Alpha diversity between the two groups(P>0.05).The difference of Beta diversity of intestinal flora between two groups was statistically significant(P<0.05).At the phylum level,Firmicutes,Actinobacteriota and Proteobacteria are the dominant phylum of the two groups,but the abundances are different.The relative abundance of Bacteroidota and Desulfobacterota in the delay period group is higher than that in the normal group(P<0.05).At the genus level,compared with the normal group,the intestinal abundances of Bacteroides were relatively abundant in the delay period group(P<0.05),while the relative abundances of fusicatenibacter were relatively low(P<0.05).LEfSe analysis showed that the abundance of bacteroides played a major role in the difference of intestinal flora in the delay period group in terms of phyla/class/order.Conclusion The intestinal microflora structure of heroin addicts is different from that of healthy people.It is considered that prolonged withdrawal symptoms may affect the intestinal microflora structure of heroin addicts.
