基于污水分析的依托咪酯滥用风险预测的方法研究

摘要
目的 针对当前麻醉药依托咪酯的滥用形势,建立基于全自动在线固相萃取分析系统-超高效液相色谱-串联质谱的分析方法,同时对城市生活污水中依托咪酯及其代谢产物依托咪酯酸进行定量检测,以预测其滥用情况.方法 将离心后的污水样品在pH 5-7条件下加入内标并过滤,利用Waters Oasis HLB column进行在线固相萃取,并采用WATERS ACQUITY UPLC HSS T3色谱柱进行色谱分析;质谱采用ESI+离子源,质谱多反应监测模式(multiple reaction monitoring,MRM)进行定量分析.结果 各目标物在0.5~100 ng/L范围内线性关系良好(r≥0.9995),日内与日间精密度均小于10.76%,基质效应与回收率均符合方法学要求.我国东部某中心城市的实际污水样品中,依托咪酯检出率为59.03%,检出浓度范围为0.522~64.338 ng/L;依托咪酯酸的检出率为40.97%,检出浓度范围为0.573~19.945 ng/L.该市下辖各区的平均依托咪酯千人均消费量范围在0.02 mg/1000inh/d~6.97 mg/1000inh/d.结论 本研究建立的方法可快速、高效、准确地检测污水中依托咪酯及其代谢物依托咪酯酸,提出了区域内依托咪酯千人均消费量的计算方法,为依托咪酯的潜在滥用风险评估提供了参考.
Abstract
Objective To establish a quantitative analysis method for the detection of etomidate and its metabolite etomidate acid in urban wastewater,aiming to predict their abuse prevalence.Methods The sewage samples were centrifuged and adjusted to pH 5-7,and Waters Oasis HLB column was used for online solid-phase extraction.The analytes were separated on a WATERS ACQUITY UPLC HSS T3 column by gradient elution and were quantitated by multiple reaction monitoring(MRM)mode with positive electrospray ionization(ESI*).Results All analytes showed good linearity(r ≥ 0.9995)in the range of 0.5-100 ng/L,the intra-day and inter-day precision were both less than 10.76%,the matrix effect and recovery rate all met the requirements of methodology validation.In the sewage samples of a central city in the east of China,the detection rate of etomidate was 59.03%,the concentration ranged from 0.522 ng/L to 64.338 ng/L;the detection rate of etomidate acid was 40.97%,the concentration ranged from 0.573 ng/L to 19.945 ng/L.The daily mass load of etomidate per 1000 inhabitants in each district of the city ranged from 0.02 mg/1000inh/d to 6.97 mg/1000inh/d.Conclusion This method can quickly,efficiently and accurately detect etomidate and its metabolite etomidate acid in sewage.The proposed calculation method for etomidate consumption per capita provides a reference for assessing the potential abuse risk associated with etomidate.
