摘要
目的 建立一种高效液相色谱-质谱联用(LC-MS/MS)法,对毛发中的喹硫平进行分段含量测定.方法 毛发样品经过冷冻研磨为粉末后用甲醇超声提取30 min,经0.22 μm滤膜.色谱柱使用Thermo Gold ODS C18(150 mm×2.1 mm,5 μm),流动相为甲醇-0.2%冰乙酸水溶液,质谱采用ESI+离子源,MRM多反应监测.结果 喹硫平在0.15 ng/mg~50 ng/mg范围内峰面积与浓度呈良好的线性关系,喹硫平的工作曲线方程为Y=1.1046X-0.9789(R2=0.9934),方法绝对回收率在83.1%~102.6%.结论 建立了分段测定毛发中喹硫平含量的LC-MS/MS法,该方法结果灵敏、准确、特异性强,在药物滥用致死的刑事案件中具有一定应用价值.
Abstract
Objective To establish a high-performance liquid chromatography-mass spectrometry(LC-MS/MS)method for the segmented determination of quetiapine in hair.Methods The hair samples were frozen and ground into power,then extracted with methanol ultrasound for 30 minutes and filtered through a 0.22 μm membrane.The chromatographic column was Thermo Gold ODS C18(150 mm ×2.1 mm,5 μm),and the mobile phase was methanol-0.2%aqueous glacial acetic acid.Mass spectrometry was performed with ESI+ion source and MRM multiple reaction monitoring.Results The peak area of quetiapine showed a good linear relationship with concentration in the range of 0.15 ng/mg to 50 ng/mg.The operating curve equation of quetiapine is Y=1.1046X-0.9789(R2=0.9934).The absolute recovery of the LC-MS/MS method was in the range of 83.1%to 102.6%.Conclusion A LC-MS/MS method for segmented determination of quetiapine content in hair has been established,which is sensitive,accurate,and highly specific.This method has certain application value in criminal cases of drug abuse causing death.
