Objective To develop a rapid and cost-effective method of high performance liquid chromatography-ultraviolet detection(HPLC-UV)for the simultaneous determination of the plasma concentrations of imipenem,meropenem and ceftazidime in human plasma.Methods The protein was removed by Millipore 10KD ultrafiltration centrifugation,and the chromatographic column was SunFire C18 column(4.6 mm×250 mm,5μm).The pre-column was C18 column(4.0 mm×2.1 mm,5μm),and the mobile phase was 0.1 mol·L-1 3-(N-morpholino)propanesulfonic acid and acetonitrile with pH 7.0.Samples were gradiently eluted with the flow rate of 1.0 mL·min-1 for 30 minutes and detected at the wavelength of 299 nm.The injection volume was 30μL.Results The retention time of imipenem,meropenem and ceftazidime were 6.699 min,10.795 min and 8.722 min,respectively.The peak form was pleasing without disturbance of the endogenous components in plasma.The method showed good accuracy,precision,and linear relationship.The samples with different concentrations showed high stability at-20℃ and 4℃.Conclusion The method has low operation cost,strong specificity and good separation effect,which can be used for routine determination of the plasma concentrations of imipenem,meropenem and ceftazidime in patients with melioidosis.
