Objective To investigate the molecular mechanism of the protective effect of Huangkui capsules on IgA nephropathy rats.Methods The model of IgA nephropathy was established by intragastric administration of serum albumin,subcutaneous injection of carbon tetrachloride and tail vein injection of lipopolysaccharide.Healthy female rats were randomly divided into control group(group A),IgA nephropathy model group(group B),Huangkui capsules treatment group(group C)and IgA nephropathy group treated with Huangkui capsules(group D).Huangkui capsules were administered to group C and group D intragastrically.Serum,urine and kidney tissue samples were collected after 4 weeks.The pathological changes of kidneys were observed.The relative mRNA expression of β-catenin and α-SMA in Wnt/β-catenin signaling pathways was detected.The protein expression of β-catenin and α-SMA was detected.The levels of urinary protein,serum creatinine and blood urea nitrogen in 24 hours were detected.Results Compared with group A,24 h kidney damage indicators such as urinary protein,serum creatinine and blood urea nitrogen,kidney tissue damage level,β-catenin and α-SMA mRNA expression,as well as β-catenin and α-SMA protein expression in group B and group D significantly increased,with statistical significance(P<0.05),that in group C showed no significant differences.Compared with group B,that in group D significantly decreased,and the differences were statistically significant(P<0.05).Conclusion Huangkui capsules had no obvious protective effect on the kidneys of healthy rats.Huangkui capsules could reduce the kidney damage and the expression of Wnt/β-catenin pathways in IgA nephropathy rats,which might be related to their inhibition of the activation of Wnt/β-catenin signaling pathways.
