Relationship between lncRNA ZFAS1,Notch1 mRNA expression in epithelial ovarian cancer tissue and 5-year survival of patients after surgery
Objective To explore the relationship between the expression levels of long non-coding RNA zinc finger protein antisense chain 1(lncRNA ZFAS1)and Notch1 messenger RNA(mRNA)in epithelial ovarian cancer(EOC)tissues and the survival of patients within 5 years after surgery.Methods A total of 96 patients with EOC(EOC group)who received surgical treatment in Handan People's Hospital from March 2015 to July 2018 were selected,EOC tissues and normal ovarian tissues adjacent to cancer were collected during the operation.The expression levels of lncRNA ZFAS1 and Notch1 mRNA in tissues were measured by real-time fluorescence quantitative PCR,patients with EOC were followed up for 5 years after surgery to record their survival.The expression levels of lncRNA ZFAS1 and Notch1 mRNA in normal ovarian tissues adjacent to cancer and EOC tissues were compared.The clinicopathological features and lncRNA ZFAS1,Notch1 mRNA expression levels of EOC tissues in survival group and death group were compared.Analyzed the correlation between the expression levels of lncRNA ZFAS1 and Notch1 mRNA in EOC tissues and their relationship with clinical pathological features and 5-year postoperative survival of patients,the risk factors affecting the 5-year postoperative survival of patients with EOC,and the predictive efficacy of lncRNA ZFAS1 and Notch1 mRNA expression levels in EOC tissues on the 5-year survival of EOC patients after surgery.Results Compared with normal ovarian tissues adjacent to cancer,the expression levels of lncRNA ZFAS1 and Notch1 mRNA in EOC tissues were increased(P<0.05);the expression levels of lncRNA ZFAS1 and Notch1 mRNA in EOC tissues were positively correlated(P<0.05),and their expression levels were not related to age,menopause or not,pathological type,or tumor size(P>0.05),they were related to FlGO staging,lymph node metastasis,and tumor differentiation degree(P<0.05);the 5-year overall survival rates of the lncRNA ZFAS1 high expression group and Notch1 mRNA high expression group were lower than those of the lncRNA ZFAS1 low expression group and Notch1 mRNA low expression group(P<0.05);there was no statistically significant difference in age,menopause or not,pathological type,and tumor size between the survival and death groups(P>0.05),but there was a significant difference in FIGO staging,lymph node metastasis,and tumor differentiation degree(P<0.05);compared with survival group,the expression levels of lncRNA ZFAS1 and Notch1 mRNA in the EOC tissues of death group were increased(P<0.05);FIGO stage Ⅲ~Ⅳ,lymph node metastasis,low tumor differentiation,high expression of lncRNA ZFAS1,and high expression of Notch1 mRNA were all independent risk factors that affect the survival of EOC patients within 5 years after surgery(P<0.05);the area under curve(AUC)of lncRNA ZFAS1,Notch1 mRNA,and their combined prediction of survival in EOC patients within 5 years after surgery were 0.861,0.913,and 0.965,respectively;the AUC predicted by the combination of the two was significantly greater than that predicted by lncRNA ZFAS1 alone and Notch1 mRNA alone(P<0.05).Conclusion The lncRNA ZFAS1 and Notch1 mRNA are highly expressed in the cancer tissues of EOC patients,and their expression is related to the survival of EOC patients within 5 years after surgery,which has a high predictive power for survival within 5 years after surgery.
