Therapeutic effect of umbilical cord blood mononuclear cells infusion on intrauterine adhesion in rats
Objective To investigate the therapeutic effect and mechanism of umbilical cord blood mononuclear cells(UCBMNCs)infusion on intrauterine adhesions(IUA)in rats.Methods 18 female SPF SD rats were randomly divided into control group,model group and UCBMNCs group.The rats in the control group were sutured immediately after laparotomy,and the rat model of intrauterine adhesions in the model group and UCBMNCs group were constructed by laparotomy curettage method.The model group was IUA model+intravenous infusion of normal saline.The UCBMNCs group was treated with intravenous infusion of UCBMNCs in the rat model of IUA.After 2 weeks of treatment,bilateral uterine tissues of each group of rats were collected to detect the number of endometrial glands,the ratio of fibrosis area,the protein and mRNA expression level of TGF-β1,MMP-9 and CTGF in endometrial tissues.Results Compared with the model group,the UCBMNCs group had a significant improvement in uterine morphology and increased endometrial glands.The percentage of endometrial fibrosis in the control group was(20.61±8.56)%,that in the model group was(54.77±10.54)%,and that in the UCBMNCs group was(35.02±14.07)%.The endometrial fibrosis of UCBMNCs group was significantly reduced(P<0.05),collagen fibrocyte infiltration could be seen in the local myometrium,but less than that in the model group.The relative expression levels of TGF-β1 in the model group and the UCBMNC group were(1.34±0.22)and(0.76±0.22),respectively,and the difference was statistically significant(t=3.228,P<0.05).The relative expression level of TGF-β1 mRNA in UCBMNCs group was(2.33±0.90),which was significantly lower than(3.55±1.29)in model group(P<0.05).Conclusions Human UCBMNCs can reduce the levels of TGF-β1 in the IUA model of rats,reduce the relative expression of TGF-β1 mRNA,inhibit endometrial fibrosis,and improve the IUA.
